Zusammenfassung
Nachdem in einer vorangegangenen NeuroMastER-Studie eine Lokalisation der mRNA des V PAC2-Rezeptors für Vasoaktives Intestinales Polypeptid (VIP) erfolgte, war es das Ziel der vorliegenden Studie, in situ Proteindaten zur Lokalisation von V PAC2 bei Bullösem Pemphigoid (BP) zu sammeln. Diese Immunerkrankung unterliegt einer Vielzahl von Umwelteinflüssen.
Mittels unterschiedlicher immunhistochemischer Verfahren, gekoppelt mit Dunkelfeld- und Hellfeld-visualisierenden Techniken, konnte dabei eine Expression von V PAC2-Immunreaktivität in folgenden Zellen von normaler und BP-Haut festgestellt werden: Keratinozyten, Zellen der Gefäßwände, Zellen ekkriner Schweißdrüsen und holokriner Talgdrüsen, Haarfollikelzellen und Zellen der glatten Muskulatur des M. erector pili. Auch Nervenfaserbündel in der Dermis zeigten vereinzelt V PAC2-Immunreaktivität.
Zusammenfassend bestätigen diese Daten auf der translationalen Ebene frühere Studien, die auf der transkriptionellen Ebene VPAC2 mRNA untersucht haben.
V PAC2, VIP und Mastzellen können als wichtige Kandidaten für die Mediation von Prozessen zwischen dem Körper und der Umwelt gelten.
Abstract
After the localisation of mRNA of the V PAC2 receptor of Vasoactive Intestinal Polypeptide (VIP) in a previous NeuroMastER-study, the present study aimed to generate in situ protein data for the localisation of V PAC2 in bullous pemphigoid (BP). This immune disease is subject to numerous environmental influences.
Using different immunohistochemical methods coupled to darkfield and brightfield visualising techniques, the expression of V PAC2 immunoreactivity was found in follwing cells of normal and BP skin: Keratinocytes, cells of vessel walls, eccrine and holocrine glandular cells, hair follicular cells, and cells of the smooth muscle of m. erector pili. Also nerve bundles of the dermis displayed immunoreactivity in part.
In summary, the present data confirm previous transcriptional data on the translational level. V PAC2, VIP and mast cells can be regarded as important candidates for the mediation of interactions between the organism and its environment.
