Abstract
Digital imaging fluorescence microscopy was used to study the effect of two antioxidants, N-acetyl-cysteine (NAC) and glutathione, on the cytosolic free calcium concentration ([Ca2+]i) induced by cholecystokinin-octapeptide (CCK-8) of mouse pancreatic acinar cells. When acinar cells were preincubated with either NAC or glutathione, subsequent stimulation with CCK-8 in the presence of each antioxidant had no significant effect on the typical pattern of [Ca2+]i transient evoked by the gastrointestinal hormone. However, application of NAC to acinar cells pretreated for 60 min with the same antioxidant, strongly blocked the oscillatory pattern initiated by CCK-8, inhibiting both amplitude and frequency of calcium oscillations. By contrast, glutathione had no effect on the oscillatory pattern evoked by CCK-8. The present results allow us to speculate that during [Ca2+]i oscillation there is a production of oxidants that facilitate oscillations by enhancing release of calcium from internal stores.
Resumen
En el presente trabajo se estudia el efecto de dos antioxidantes, N-acetil-cisteína (NAC) y glutation, sobre la concentración de calcio libre citosólico ([Ca2+]i) inducido por colecistoquinina-octapéptido (CCK-8) en células acinares pancreáticas de ratón mediante el uso de un microscopio de fluorescencia. Cuando las células acinares se preincuban en presencia de NAC o glutation, la posterior estimulación con CCK en presencia del respectivo antioxidante no modifica significativamente la típica respuesta en la [Ca2+]i evocada por la hormona gastrointestinal. Sin embargo, la aplicación de NAC a células acinares pretratadas durante 60 minutos con el mismo antioxidante, bloquea el patrón oscilatorio iniciado por la CCK-8, inhibiendo tanto la amplitud como la frecuencia de las oscilaciones. Por el contrario, el glutation se muestra inefectivo sobre el patrón oscilatorio evocado por CCK. Estos resultados nos permiten especular la producción de agentes oxidantes facilitando las oscilaciones de calcio por aumentos en la liberación de calcio desde depósitos internos.
