Abstract
Nitric oxide (NO) can stimulate dendritic cells to a more activated state. However, nitric oxide and peroxynitrites production by dendritic cells has been usually associated with pathological situations such as autoimmunity or inflammatory diseases. This study was designed to determine if dendritic cells obtained from healthy volunteers produce nitric oxide and peroxynitrites, which results in protein nitration. The expression of arginase II, but not arginase I, isoform was detected in monocytes and dendritic cells. There was higher inducible nitric oxide synthase (iNOS) protein expression and lower arginase activity both in immature and mature dendritic cells, compared to monocytes. This caused nitric oxide production, and maturation of dendritic cells which provoked a significative increase of nitrites and nitrates compared to immature dendritic cells. There was also peroxynitrites synthesis during monocyte differentiation as shown by the nitration of proteins. Immunoblot revealed a pattern of nitrated proteins in cell extracts obtained from monocytes and dendritic cells, however there were bands that appeared only in human dendritic cells, in particular an intense 90 kDa band. Nitric oxide production and nitrotyrosine formation could affect the antigen presentation and modify the immune response.
Resumen
El óxido nítrico (NO) puede estimular a las células dendríticas hacia un estadío más activado. Asimismo, la producción de óxido nítrico y de peroxinitritos por las células dendríticas se ha asociado comúnmente a diversas situaciones patológicas como la autoinmunidad o procesos de inflamación. Este estudio fue diseñado para determinar si las células dendríticas obtenidas de voluntarios sanos producen óxido nítrico y peroxinitritos, implicados en la nitración de proteínas. La expresión de la isoforma de arginasa II, pero no de arginasa I, fue detectada en monocitos y en células dendríticas. Se observó una elevada expresión de la isoforma inducible de la óxido nítrico sintasa a nivel de proteína y una menor actividad arginasa en las células dendríticas inmaduras y maduras, en comparación con la detectada en monocitos. Este balance dió lugar a la producción de óxido nítrico, y la maduración de las células dendríticas produjo un incremento significativo de los nitritos y nitratos comparado con las células dendríticas inmaduras. Durante la diferenciación de monocitos se produjo síntesis de peroxinitritos como mostró la nitración de proteínas. El patrón de nitración de proteínas, que se estudió en los extractos celulares mediante Western blot, resultó similar en monocitos y en células dendríticas, aunque se observó una banda de 90 kDa que aparece sólo en células dendríticas y no aparece en los monocitos. La producción de óxido nítrico y la formación de nitrotirosina podría afectar a la presentación antigénica y modificar la respuesta inmunológica.
