Zusammenfassung
Einleitung
Bei Verätzungen des Auges durch Säuren oder Laugen können auch Bindehautzellen geschädigt werden. Hier wird eine Methode beschrieben, mit der die Mitochondrienaktivität humaner Bindehautzellen gemessen werden kann. Damit ist es möglich, das Ausmaß der Schädigung der Bindehautzellen nach Verätzung festzustellen.
Methode
Humane Bindehautzellen (Chang-Zellen) werden kultiviert und mit Säure (0,02 M; 0,05 M; 0,1 M HC1) oder Lauge (0,02 M; 0,05 M; 0,1 M NaOH) eine definierte Zeit (10–60 sec) inkubiert. Zur Feststellung der Mitochondrienaktivität wird der Cell-Titer 96® Aqueous Solution Cell Proliferation Assay verwendet.
Ergebnisse
Innerhalb von wenigen Sekunden kommt es durch Säure oder Lauge in Abhängigkeit von deren Konzentration zu einer Abnahme der Mitochondrienaktivität von Bindehautzellen.
Schlussfolgerung
Diese Methode erlaubt es, die Schädigung der Bindehautzellen durch Säuren oder Laugen quantitativ zu messen. Damit kann man auch die Wirksamkeit von Substanzen untersuchen, die die geschädigten Zellen nach Verätzungen wieder regenerieren sollen.

Introduction
Chemical burns of the eye by acid or alkali may also damage conjunctival cells. We describe a method to measure the mitochondrial activity of human conjunctival cells. By this method the degree of injury to the cells by chemical agents can be determined.
Method
Human conjunctival cells (Chang cells) in cell culture were incubated with hydrochloric acid (0.02 M; 0.05 M; 0.1 M HC1) or sodium hydroxide solution (0.02 M; 0.05 M; 0.1 M NaOH) for a defined period of time (10–60 sec). To determine the mitochondrial activity of the cells the Cell Titer 96® Aqueous Solution Cell Proliferation Assay was used.
Results
Within few seconds the mitochondrial activity of the conjunctival cells decreased depending on the concentration of the acidic or alkaline solution and on the time of incubation.
Conclusion
By this method the damage of human conjunctival cells by acid or alkali can be determined quantitatively. Evaluation of the activity of substances for cell regeneration after chemical burns is also possible.
