Zusammenfassung
Hintergrund
Zymographie Ergebnisse deuteten auf die Bedeutung der Matrix Metalloproteinasen (MMP) und deren Inhibitoren (TIMPs) in der Entwicklung des Keratokonus hin. Diese Arbeit untersucht die Lokalisation von MMP-2, MMP-3, MMP-9, TIMP-1 und TIMP-2 in normalen Hornhäuten und jenen mit Keratokonus.
Material und Methoden
Zehn Hornhäute mit Keratokonus und acht normale Hornhäute (in Paraffin eingebettet) wurden mit Hilfe der Streptavidin-Biotin Peroxidase Technik auf MMP-2, MMP-3, MMP-9, TIMP-1 und TIMP-2 gefärbt.
Ergebnisse
MMP-2 und MMP-3 wurden ausschließlich im kornealen Epithel gefunden und das gesamte Stroma und Endothel zeigten keinerlei Reaktionen auf diese beiden MMPs. Dabei gab es keinen Unterschied zwischen normalen Hornhäuten und jenen mit Keratokonus. MMP-9 konnte in normalen Hornhäuten nicht gefunden werden, war aber in Korneas mit Keratokonus an wenigen Stellen im zentralen kornealen Epithel darstellbar. TIMP-1 und TIMP-2 waren im Epithel und im Stroma zu finden und es zeigte sich keine unterschiedliche Expression zwischen normalen Hornhäuten und jenen mit Keratokonus.
Schlussfolgerungen
MMP-2, MMP-3 und MMP-9 konnten zwar im kornealen Epithel, nicht jedoch im Stroma oder im Endothel nachgewiesen werden. Auch die Expression der TIMPs in normalen Hornhäuten zeigte keinen Unterschied zu Hornhäuten mit Keratokonus. Diese Ergebnisse deuten darauf hin, dass MMP-2, MMP-3 und MMP-9 und deren Inhibitoren TIMP-1 und TIMP-2 bei der Entwicklung des Keratokonus keine Rolle spielen.
Summary
Background
Zymography results indicated a role of matrix metalloproteinases (MMPs) and their inhibitors (TIMPs) in the development of keratoconus. This study investigated the localisation of MMP-2, MMP-3 and MMP-9, TIMP-1 and TIMP-2 in keratoconus tissues.
Materials and Methods
Ten corneas with keratoconus and eight normal corneas (paraffin embedded) were stained for MMP-2, MMP-3 and MMP-9, TIMP-1 and TIMP-2 by the labled streptavidin-biotin peroxidase technique.
Results
MMP-2 and MMP-3 could only be found in the corneal epithelium, but the stroma and the endothelium lacked expression of those two MMPs. No difference could be observed between normal tissues and those with keratoconus. MMP-9 was negative in the entire normal cornea, but was found in few small areas of the central corneal epithelium in cases of keratoconus. TIMP-1 and TIMP-2 were positive in the epithelium and stroma, but no difference was observed between normal and keratoconus corneas.
Conclusions
MMP-2, MMP-3 and MMP-9 were detected in the corneal epithelium, but not in the corneal stroma or in the endothelium. There was no difference in the expression of TIMP-1 and TIMP-2 between normal and keratoconus corneas. This indicates, that MMP-2, MMP-3 and MMP-9 and their inhibitors TIMP-1 and TIMP-2 play no role in the development of keratoconus.
