Abstract
De ziekte van Parkinson wordt veroorzaakt door het afsterven van dopamine producerende zenuwcellen in de middenhersenen. Bekend is dat oxidatieve stress, optredend door een disbalans tussen aanmaak van giftige zuurstofverbindingen en de ontgifting daarvan, een hoofdrol vervult in het neurodegeneratieve proces. Curieus genoeg blijkt het metabolisme van dopamine zelf een belangrijke bron te zijn van oxidatieve stress. Een van de mogelijkheden om deze oxidatieve stress te bestrijden is het stimuleren van de activiteit van enzymen die toxische dopaminemetabolieten onschadelijk maken. Of farmaca met een dergelijke ontgiftingsinducerende werking ook daadwerkelijk neuroprotectief zijn, wordt momenteel onderzocht binnen het ‘Parkinsonprogramma’ van het vumc te Amsterdam.
