Summary
Alpha-1-antitrypsin deficiency is an autosomal recessive disease. The prevalence of the disease is the utmost among the white North European population. The clinical picture is very variable and can range from mild liver enzyme elevation in early years of life to liver cirrhosis. In adults the disease can present with lung emphysema. The liverdamage is caused by a folding defect from the mutated α1-antitrypsine resulting in liver storage. Lung emphysema is caused by α1-antitrypsine deficiency by the absence of the protease inhibitor activity of α1-antitrypsine resulting in lungdestruction. We will discuss genetic aspects, pathophysiology, the clinical picture, diagnostic procedures, treatment, and the advantages and disadvantages of screening α1-antitrypsine deficiency.
