Abstract
Reaction of tetragonal hen egg-white lysozyme with excess iodine in potassium iodide in 0·02M acetate buffer at pH 4·7 gave the heavy atom derivative : iodolysozyme which has been crystallized in the same form.
Tryptic digestion of heat denatured iodolysozyme and paper electrophoresis, at pH 3·6 and pH 6·4 of the digest have shown that only the single histidine in lysozyme is iodinated. This location by chemical means, of the particular amino-acid residues attacked by I+ in lysozyme, is in disagreement with the crystallographic location by Blake who has investigated the reaction of tetragonal lysozyme with iodine crystallographically.
Iodolysozyme was found to be about 80 per cent biologically active against the cell-walls ofMicrococcus lysodeikticus.
