Zusammenfassung
Im Rahmen der Begleitforschung zur Freisetzung von transgenem Raps (Brassica napus) werden Auskreuzungshäufigkeit, Vektoren und Reichweite der Ausbreitung der Herbizidresistenz (Phosphinothricin-Acetyltransferase-Gen:pat-Gen) auf Akzeptorpflanzen von nicht-transgenem Raps und Rübsen (Brassica rapa) untersucht. Der im Sommer 1997 freigesetzte transgene Raps war von einer Mantelsaat aus nichttransgenem Raps umgeben. Konzentrisch um das Versuchsfeld wurden an 35 Punkten in den Abständen 50 m, 75m, 100 m, 150 m, 210 m, 300 m und 400 m jeweils 100 getopfte Pflanzen von Raps und Rübsen ausgebracht. Diese konnten in der Blüte mit den Pflanzen im Versuchsfeld synchronisiert werden. Vor Erreichen der Vollreife wurden die Samen von Raps und Rübsen geerntet. Aus der Mantelsaat wurde an sechs Punkten ebenfalls Saatgut gewonnen. Derzeit werden Samen aus diesen Probepunkten im Gewächshaus angekeimt und mit dem Totalherbizid Basta behandelt. In den überlebenden Pflanzen konnte mit Hilfe der PCR daspat-Gen quantitativ nachgewiesen werden. Der Anteil der resistenten Pflanzen schwankt von Punkt zu Punkt. In der Mantelsaat liegen die Werte zwischen 0,3% und 2,2%. Bei Samen, die aus 100 m Entfernung stammen, sind noch 0,5% der gekeimten Pflanzen Basta-resistent.
Die Blütenbesucher im Versuchsfeld und auf den Akzeptorpflanzen wurden mit verschiedenen Fangmethoden (Kescherfang, Gelbschalen, Malaisefallen) erfaßt. In den bislang ausgewerteten Kescherfängen ergab sich eine nennenswerte Pollenfracht bei Haut- und Zweiflüglern. Wildbienen und Schwebfliegen waren häufiger anzutreffen als Honigbienen und Hummeln. Im Pollen, der von den Tieren stammte, konnte daspat-Gen ebenfalls nachgewiesen werden.
Summary
During field trials with transgenic rape (Brassica napus) the frequency of outcrossing, the vectors and the dissemination distance of the herbicide-resistance (phosphinothricineacetyltransferase-gene:pat-gene) on to acceptor plants of non-transgenicBrassica napus orBrassica rapa were investigated. The field trial with transgenic rape plants were surrounded by a belt of non-transgenic rape plants. In the distances of 50 m, 75 m, 100 m, 150 m, 210 m, 300 m and 400 m from the field trial non-transgenic plants ofBrassica napus andBrassica rapa were exposed as acceptor plants. The outcrossing rates of the transgene were determined by Basta application and controlled by PCR (between 0.3% and 2.2% in the belt of non-transgenic rape plants and 0.5% in a distance of 100 m from the field trial).
The pollinators within the area of the field trial and on the acceptor plants in the surrounding were captured by different methods. Most of the captered pollinators have collected remarkable amounts of transgenic pollen.
