Optimized production of a thermostable lipase by recombinantPseudomonas putida 1641

Abstract

A thermostable lipase gene fromPseudomonas fluorescence was cloned into a multicopy plasmid and electroporated intoP. putida. The recombinantP. putida produced soluble lipase into the broth. Using these recombinant cells, the optimum conditions for lipase production were investigated. Olive oil was the most effective carbon source, producing upto 60 unit/ml. Supplementation of the medium with a surfactant, such as Tween-80, increased the lipase productivity significantly. The optimum temperature and pH for lipase production were 26°C and 7.0, respectively.
