Abstract
V listech pšenice, pěstované 4 až 10 dní v roztoku kys. giberelové (10–20 p.p.m.) bylo chromatografickou analysou zjištêno zvýšení hladiny fruktoso-1,6-difosfátu v poměru ke kyselině 3-fosfoglycerové a fosfoenolpyrohroznové a kyseliny 3-fosfo-glycerové v poměru k dosud neidentifikovanému fosfátu. Tyto změny byly nalezeny za různých světelných režimů, jež předcházely extrakci fosfátů z listů. Na základě těchto údajů byl se zřetelem k výsledkům dřívějších pokusů (Luštinec, Pokorná aRůžička 1962) vysloven předpoklad o interakei mezi glykolysou a reakcemi spotřebovávajícími její meziprodukty pro synthesu lipidů jakožto o způsobu, kterým se může měnit působením kyseliny giberelové hladina analysovaných fosfátů v listech. Za účelem ověření jeho správnosti byla sledována inkorporace 1-14C- a 6-14C-glukosy do lipidů, fosfolipidů (jejichž hořečnatá sůl je nerozpustná v acetonu), CO2 a nelipidické frakce listů. Ukázalo se, že inkorporace 1-14C do lipidických frakcí rostlin pěstovaných 3 až 7 dní v roztoku kys. giberelové je nápadně snížena ve srovnání s inkorporací 6-14C. Poměr radioaktivit C6∶C1 byl v lipidech značně zvýšen, také tříhodinovým působením kyseliny giberelové (10 a 50 p.p.m.) na listy při jejich inkubaci s glukosou-14C. Vzhledem k tomu, že množství látek extrahovatelných etherem je v listech pokusných rostlin stejné jako u kontroly a že inkorporace 1-C do lipidů je v řadě případů u kontrolních rostlin značně vyšší než inkorporace 6-C, je nutno uvažovat o účinku kyseliny giberelové na rychlost výměny uhlíkových atomů mezi lipidy a produkty katabolismu glukosy.
Summary
Chromatographic analysis of wheat leaves grown for 4 to 10 days in a solution of gibberellic acid (10–20 p.p.m.) revealed a higher level of fructose-1,6-diphosphate as compared with that of 3-phosphoglyceric acid and phosphoenolpyruvic acid, and of 3-phosphoglyceric acid as compared with that of a hitherto unidentified phosphate. These changes were found under different light regimes which preceded extraction of phosphates from leaves. On the basis of these data and in view of earlier results (Luštinec, Pokorná, Růžička 1962) it is assumed that glycolysis interacts with reactions utilizing its intermediate products for the synthesis of lipids, this representing a possibility of bringing about changes in leaf phosphate levels due to gibberellic acid. In order to verify the postulate the incorporation of 1-14C- and 6-14C-glucose into lipids, phospholipids (the magnesium salts of which are insoluble in acetone), carbon dioxide and the non-lipid fraction of leaves was investigated. It appeared that the incorporation of 1-14C into the lipid fractions of plants grown for 3–7 days in a gibberellic acid solution is markedly lower than that of 6-14C. The ratio of radioactivities C6∶C1 was increased in the lipids by the three-hour treatment of leaves with gibberellic acid (10 and 50 p.p.m.) during their incubation with glucose-14C. In view of the fact that the amount of substances extractable with ether is the same in the experimental and in the control leaves and that the incorporation of 1-C into lipids is frequently considerably higher in the controls than the incorporation of 6-C the effect of gibberellic acid on the rate of exchange of carbon atoms between the lipids and the products of glucose catabolism should be borne in mind.
Abstract
В листьях пшеницы, выращиваемой 4–10 дней в растворе гиббереллиновой кислоты (10–20 p.p.m.) было обпаружено с помощью хроматографического анализа повышение уровня фруктозо-1,6-дифосфата по отношению к 3-фосфоглицериновой и фосфоэнолнировиноградной кислогам и 3-фосфоглицериновой кислоты по отношению к неопределенному фосфату. Эти изменения были обнаружены при различных световых режимах, которые предшествовали экстракции фосфатов из листьев. Исходя из этих данных и с учетом раньше полученных результатов (Luštinec, Pokorná аRůžička 1962) было высказано предноложение о взаимовлиянии между гликолизом и реакциями потребляющими его межнродукты для синтеза липидов как о способе, с помощью которого изменяется уровень апализированных фосфатов в листьях благодаря действию гиббереллиновой кислоты. С целью проверки правильности этого предположения изучалась инкорпорация 1-14C- и 6-14Сглюкозы в липиды, фосфолипиды (магниевая соль которых нерастворима в ацетоне), CO2 и нелипидические фракции листьев. Было найдено, что инкорпорация 1-14C в липидические фракции растений выращиваемых 3–7 дней в растворе гиббереллиновой кислоты выразительно снижается по сравнению с инкорпорацией 6-14C. Отношение радиоактивностей C6/C1 было в липидах намного побышено благодаря З часовому действию гиббереллиновой кислоты (10 и 50 p.p.m.) на листья во время их инкубации с глюкозой C14.
В связи с тем, что количество вещесть экстрагируемых эфиром является одинаковым в листьях подопытных растений и контроля, и что инкорпорация 1—С в липиды в ряде случаев у контрольных растений значительпо выше чем инкорпорация 6—С, необходимо предполагать действие гиббереллиновой кислоты на скорость обмена углеродных атомов между линидами и продуктами катаболизма глюкозы.
