Abstract
We compared the occurrence of peroxidase isozymes in protein extract from roots, hypocotyls and cotyledons of 10 dayCucurbita pepo plants and of adult leaves of older plants by means of starch gel and polyacryl amide gel electrophoresis. We reached maximum discrimination by means of starch gel electrophoresis: 11 zones were ascertained on the cathode side and about 2 on the anode side at pH 3.1. Two zones occurred regularly:A and (the latter having a more complicated structure). ZoneD is characteristic for roots, but is it suppressed and seldom found with leaves. On the other hand zonesC 1 andC 2 are clearly discernible with leaves but are substantially less evident with roots. The character of anodic zoneZ is discussed later in this paper.
Abstract
Srovnávali jsme pomocí škrobové nebo polyakrylamidové elektroforézy přítomnost různých peroxidáz v bílkovinných extraktech z kořenů, hypokotylů a děloh desetidenních rostlin tykve (Cucurbita pepo L.) a z listů starších rostlin. Optimální rozlišení jsme získali pomocí škrobové elektroforézy při pH = 3.1; 11 zón na anodické a cca 3 na katodické straně. 2 zóny se vyskytovaly pravidelně, A(druhá se složtější strukturou). Zóna D je charakteristická pro kořeny, zatímco v listech bývá značně potlačena nebo není průkazná. Naproti tomu zóny C1 a C2 jsou velmi výrazné u listů a podstatně méné zřetelné v kořenech. Charakter anodické zóny Z je předmětem širší diskuse v předložené práci.
Abstract
При помощи электрофореза на крахмальном геле (или полиакриламидном геле) мы сравнивали паличие разнокачественных пероксидаз в белковьгх вытяжках из корней, гинокотилей и семядолей растении тыквы (Cucurbita pepo) в возрасте 10 дней, и из листьев взрослых растений. Самое хорошое разделепие ферментов мы получили на крахмальном геле, где появилось 11 зон в катодной части и приблизителъно три в анодной при pH = 3,1.
Мы всегда встречали две зоны: A и B (вторая с более сложным строением). Для набора пероксидаз корней характерным компонентом является D, мало или вовсе пе появляющийся в листовых органах. Б листьях, наоборот, ферменты C1 и C2 очень четкие, которые, однако, мало заметныв корнях. Б работе также обсуждается природа анодной зоны Z.
