Summary
Salmonella paratyphi endotoxin was prepared possessing the pyrogenic and biological manifestations described in the literature. A method was developed of administering the endotoxin into the inner cerebrospinal passages of the brain and a new technique of studying the state of thermoregulation, by means of hyperaemization latent period of the rabbit ear-lobe.
It was found that on intracerebral administration the effect of endotoxin was a hundred to a thousand-fold higher, as compared with intravenous administration. The type of fever reaction and its latent period are essentially the same on intracerebral as on intravenous endotoxin administration. The extent of the fever reaction with intracerebral administration of endotoxin is a function of the local concentration of endotoxin.
On the basis of these findings the authors presume that endotoxin probably increases the temperature through a direct effect on the brain tissue. But its effect is not reflex in the neuro-physiological sense of the word; it is probably a quantitatively different, much slower action. Tissue products — mediators — arising during this process alter the irritability of the thermoregulatory centre.
