Histochemical investigation of dehydrogenases in shoot apices of wheat plants at different ontogenetic stages

Histochemické studium dehydrogenas ve vzrostných vrcholech pšenice

Abstract

The activity and localization of alcohol (AD), lactic (LD), malic (MD), isocitric (ICD) and succinic (SD) dehydrogenase, resp. was histochemically determined in shoot apices of plants in the vegetative condition during transition to flowering and at the reproductive state. The enzymes were determined in freehand sections, as well as in shoot apices incubatedin toto according toNachlas et al. (1958) using Nitro-BT. The characteristic localization of the enzymes depending on the ontogenic stage of the shoot apex is described. Different parts, layers and cell groups of the apex showed differences in enzyme activity. SD could not be detected in shoot apices. The histochemical findings are discussed in relation to former results obtained from a biochemical investigation of respiration in the same material.

Abstract

Ve vzrostných vrcholech rostlin vegetativních, přecházejících ke kvetení a generativních byla histochemicky sledována aktivita a lokalisace dehydrogenasy alkoholu (AD), kyseliny mléčné (LD), kyseliny jablečné (MD), kyseliny isocitronové (ICD) a kyseliny jantarové (SD), (v ručních řezech a vzrostných vrcholech inkubovaných in toto metodou podleNachlase a spol. 1958 s Nitro-BT). Byla popsána charakteristická lokalisace enzymů související s určitou ontogenetickou etapou vzrostného vreholu: vysoká aktivita LD u juvenilních vzrostných vrcholů, vysoká aktivita AD, ICD a LD ve skupině terminálních iniciál vegetativních vrcholů, změna v lokalisaci AD v základech klásků generativních vrcholů oproti lokalisaci v mladších pupenech. Z hlediska diferenciace meristematických buněk pletiva vzrostného vrcholu byly zjištěny některé rozdíly v aktivitě sledovaných enzymů v různých částech, vrstvách, resp. skupinách buněk. SD nebyla ve vzrostných vrcholech prokázána vůbec. Histochemické nálezy jsou diskutovány v souvislosti s dřívějšími výsledky biochemického studia respirace téhož materiálu.
