Abstract
A study was performed on the influence of obligate parasitesErysiphe graminis Dc. andPuccinia hordei Ott4 on peroxidase activity in barley leaves, namely in both susceptible and resistant varieties. Attention was also paid to peroxidase activity in a susceptible wheat variety infected withPuccinia striiformis West.
The following results were obtained:
In leaves of wheat and harley plants an enzyme was revealed which oxidized pyrocatechol and guajacol only in the presence of hydrogen peroxide. This enzyme can be inactivated by heating and strongly inhibited by cyanide. Evidently peroxidase is concerned.
Peroxidase in susceptible varieties of wheat and barley is markedly activated after infection with parasites mentioned above, this activation reaching maximum at the periode of parasite sporulation. On the other hand the barley varieties resistant toE. graminis andP. hordei, exhibit only a slight or no increase in peroxidase activity at this period. Thus we come to the conclusion that peroxidase does not probably appear to be the cause of the resistance of these varieties. It seems that in tissues attacked by obligate-parasites the intensity of its activation can be a result of the infection intensity.
Abstract
Byl studován vliv obligátních parazitůErysiphe graminis Dc. aPuccinia hordei Otth na aktivitu peroxidázy v listech ječmene a to u náchylných i odolných odrůd. Pozornost byla též věnována aktivitě peroxidázy v náchylné odrůdě pšenice, infikované rzí plevovouPuccinia striiformis West.
Bylo dosaženo těchto výsledků:
V listech pšenice a ječmene byl zjištěn enzym, který oxiduje pyrokatechol a guajakol jen v přítomnosti peroxidu vodíku. Tento enzym je inaktivován teplem a silně inhibován kyanidem. Jedná se tedy o peroxidázu.
Peroxidáza náchylných odrůd pšenice i ječmene se po infekei výše uvedenými parazity význačně aktivuje a její aktivace dosahuje maxima v období sporulace parazitů. Naproti tomu odrůdy ječmene, odolné kE. graminis aP. hordei, vykazují v tomto období jen slabé nebo žádné zvýšení peroxidázové aktivity. Docházíme proto k názoru, že peroxidáza není pravděpodobně příčinou odolnosti těchto odrůd. Zdá se, že u tkání, infikovaných obligátními parazity, může být intenzita její aktivace výsledkem intenzity napadení.
