Abstract
Changes in the protein constituents in pea stem sections during root formation and its inhibition by kinetin and ethionine were studied. Only quantitative differences in the protein fractions separated on DEAE-cellulose column were noted.
The formation of foci of meristematic cells in pericycle was accompanied by an increase in the amount of fraction “l”. This fraction disappeared rapidly in sections where root formation either did not occur (internodial sections without nodes) or was inhibited by ethionine (stem sections with basal and apical nodes). Incubation of stem sections in a kinetin solution for 16 hours after cutting of stems partially preserved fraction “l”. The increase in the amount of fraction “l” was one of the first metabolic changes in root-forming pea stem sections after cutting of stems.
Abstract
Byly sledovány změny v bílkovinných frakcích oddělených chromatografií na DEAE celulóze v úsecích stonků hrachu v průběhu tvorby adventivních kořenů a při její inhibici vlivem kinetinu a ethioninu. Na chromatogramech byly zjištěny pouze kvantitativní změny v zastoupení jednotlivých frakeí. Při tvorbě meristematických ohnisek v oblasti pericyklu se v úsecích značně zvyšovalo relativní zastoupení frakce „l”. Zastoupení této frakce se značně snižovalo v průběhu kultivace úseků, ve kterých se nevytvářely adventivní kořeny (úseky bez uzlin a úseky inkubované v roztoku etioninu). Tento pokles byl částečně omezen působením kinetinu. Relativní zvýšení obsahu frakce „l” v úsecích stonků hrachu při tvorbě meristematických ohnisek bylo jednou z prvních změn v metabolismu, která byla zaznamenána po oddělení úseků.
Abstract
Изучались изменения в белковых фракциях полученных хроматографированием на DEAE целлюлозе в сегментах стебля гороха в течение процесса образования придаточных корней и при ингибировании этого процесса кинетином и этионином. На хроматограммах установлены лишь количественные изменения в отдельных фракциях. При образовании меристематических центров в области перицикла в сегментах значительно повышалась доля фракции «г». холичество этой фракции значительно снижалось при культивации сегментов, в которых не образовывались придаточные корни (сегменты без узлов и сегменты инкубированные в растворе этионина). Это снижение было частично остановлено кинетином. Относительное повышение содержания фракции «г» в сегментах стебля тороха при образовании меристематических центров было одним из первых изменений обмена, отмеченных после отделения сегментов.
