Summary

After pretreatment with verapamil (Ver) and tetrandrine (Tet), the amount of ouabain required to induce the onset and peaking of inotropic effect as well as toxic reaction was increased, while peak value of positive inotropic effect was unaltered.

Neither the toxicity of ouabain nor its margin of safety was changed, but preventive and therapeutic action of Tet and Ver on ouabain-induced arrhythmia were demonstrated.

The anti-arrhythmic effects of tetrandrine, like Ver, may be relevant to Ca-antagonistic action.

Nach Vorbehandlung mit Verapamil (Ver) und Tetrandrin (Tet) war die Menge an Ouabain, die nötig war, um sowohl eine maximale inotrope Wirkung als auch eine toxische Reaktion zu erzielen, erhöht, der Maximalwert der positiv inotropen Wirkung war dagegen unverändert. Weder die Toxizität noch die Sicherbeitsbreite von Ouabain waren verändert, jedoch wurde präventive und therapeutische Wirkung von Tet und Ver auf die Ouabain-induzierte Arrhythmie nachgewiesen. Diese antiarrhythmische Wirkung des dem Tet ähnlichen Ver könnte für dessen Calcium-antagonistische Wirkung von Bedeutung sein.
