Abstract

A subtraction hybridization procedure using magnetic oligo(dT) particles originally designed for mRNA isolation was developed. Magnetic particles are used to isolate mRNA, prime cDNA synthesis, and eliminate transcripts common to control and experimental samples. The protocol was utilized to isolate 2 tomato cDNAs upregulated during infection withVerticillium dahliae race II.
