Zusammenfassung
Die Anwendung von enzymatisch-optischen Methoden machte es möglich, im Extrakt der Hornhaut eines Kaninchens 13 Metabolite des Energie liefernden Stoffwechsels zu bestimmen. Neben den in vivo-Gehalten der frischen normalen Hornhaut untersuchten wir auch isolierte Hornhäute nach Lagerung in einer feuchten Kammer und in den verschiedenen Stadien der Eintrübung. Der zeitliche Ablauf der Hornhauttrübung wurde über 120 Std photometrisch registriert. In Gegenwart von Luftsauerstoff nahm in der isolierten Hornhaut der Glucosegehalt sehr rasch ab. Nach 30 min war keine Glucose mehr nachweisbar. Die Gehalte von Lactat und Pyruvat fielen kontinuierlich und erreichten nach 120 Std den Nullpunkt. Die energiereichen Phosphatverbindungen ATP, ADP und AMP veränderten sich nur wenig, solange die Hornhaut klar war. Kurz vor der sichtbaren Eintrübung nach 50 Std Lagerung nahm ihr Gehalt stärker ab. Die Lactat/Pyruvat-Quotienten blieben während der ganzen Zeit nahezu konstant. Das Verhältnis ATP/ADP lag 50–60 Std lang genau so hoch wie bei frischen in vivo analysierten Hornhäuten und fiel steil ab, wenn die Trübungskurve anstieg. Dabei zeigte sich eine enge Beziehung zwischen den ATP/ADP-Quotienten und der Trübung der isolierten Hornhaut. Die langdauernde Konstanz der Lactat/Pyruvat- und der ATP/ADP-Quotienten ließ auf einen annähernd normalen Oxydationsstoffwechsel in der unter den beschriebenen Bedingungen isolierten Hornhaut schließen.
Summary
By the application of enzymatic-optic methods it was possible to estimate the levels of 13 metabolites of the energy producing metabolism in the total rabbit cornea. Besides the in vivo levels of normal fresh corneas we investigated isolated corneas stored in humid air at different states of turpidity. The develoment of turpidity with time was measured by photometry. In O2 containing atmosphere the glucose disappeared very rapidly. 30 minutes later glucose was no longer to be found. The levels of lactate and pyruvate dropped continuously and were nearly disappeared 120 hours later. The energy rich phosphate compounds ATP, ADP, AMP did not alterate much, as long as the cornea was clear. A short time befor visible turpidity, after 50 hours storage, these metabolites decreased rapidly. The lactate/pyruvate quotients were constant over a long period of time. The relation of ATP/ADP remained over a period of 50 hours as high as in the normal corneas analysed in the in vivo state, and dropped very rapidly when the curve of turpidity rose. There was a close relation between ATP/ADP-quotient and turpidity of the isolated cornea.
The long period of constant lactate/pyruvate- and ATP/ADP-quotients lead to the conclusion, that the oxydative metabolism in the isolated cornea was almost normal, if it was stored as in these experiments.
