Fluorescence of glycyl peptides and collagen degradation mixtures

Abstract

The fluorescence curves of partial acid hydrolysates of dentin and skin collagens were compared with the curves of aqueous solutions of 21 amino acids, and 16 small peptides which might be obtained from the backbone chain of collagen. The collagen degradation mixtures showed maximum emission at 410 mμ. Of the amino acids, only hydroxylysine had an emission peak at 410 mμ. Glycyl peptides in which glycine was amino-terminal also gave maximum emission in this region. Such peptides, present in high concentration in collagen hydrolysates, may account for much of the observed fluorescence.

Résumé

Les courbes de fluorescence d’hydrolysats partiels acides de collagène dentinaire et cutané sont comparées avec celles de solutions aqueuses de 21 amino-acides et 16 petits peptides suseptiblas d’être isolés de la chaîne puncipale. Les mélanges de collagène dégradé présentent un maximum d’émission à 410 mμ. De tous les amino-acides, seul l’hydroxylysine présente un pic d’émission à 410 mμ. Les peptides de glycyle, ayant la glycine en position aminoterminale, donnent une émission maximale dans cette région. Ces peptides, présents en concentration élevée dans les hydrolysats de collagène, pourraient provoquer la majeure partie de la fluorescence observée.

Zusammenfassung

Die Fluorescenzkurven von teilweise sauren Hydrolysaten von Dentin- und Hautcollagen wurden verglichen mit den Kurven von wässerigen Lösungen von 21 Aminosäuren und von 16 kleinen Peptiden, welche in der Gerüstkette von Collagen gefunden werden können. Das degradierte Collagen zeigte ein Emissionsmaximum bei 410 mμ. Bei den Aminosäuren hatte nur Hydroxylysin ein Emissionsmaximum bei 410 mμ. Die Glycylpeptide, bei welchen Glycin amino-endständig ist, zeigten ebenfalls das Emissionsmaximum in diesem Bereich. Solche Peptide, welche in hoher Konzentration in den Collagenhydrolysaten vorhanden sind, machen wahrscheinlich einen großen Teil der beobachteten Fluorescenz aus.
