Optimization of an HPLC method for the separation of erythromycin and related compounds using factorial design

Summary

An HPLC method based on a prodigy ODS2 column with a mobile phase contianging phosphate buffer, tetrabutylammonium hydrogen sulfate (TBA) and an acetonitrile gradient was optimized for the determination of erythromycin in delayed-release capsules. A central composite face-centered (CCF) factorial design was used with the factors pH, concentration of TBA and gradient slope. The method could separate erythromycin A from 7 related erythromycins.
