Abstract
Considerable amounts of orcinol-reacting material other than RNA-ribose were extracted with hot PCA from isolated spinach and broad bean chloroplasts, while cold PCA only incompletely extracted the chloroplast RNA. The procedure utilizing alkaline hydrolysis followed by an orcinol test was found to be appropriate for measuring the chloroplast RNA.
