Abstract
Activity of cytochrome P-450 (cyt. P450) is often measured to evaluate the function of liver cells in bioartificial liver. Lidocaine is often employed as a substrate, and the amount of lidocaine remaining in the medium is analyzed by either gas chromatography (GC) or highperformance liquid chromatography (HPLC) after extraction. By GC analysis, the recovery of lidocaine was about 85% after extraction. This result suggests that the activity of cyt. P450 is estimated to be higher than the actual level if such a loss in recovery is not considered. It was also observed that albumin in the medium seriously affected the cyt. P450 activity assay, when lidocaine was at a high concentration in the medium. These points should be noted in the study of bioartificial liver. It was also shown that the extraction problem mentioned above could be overcome by directly injecting medium onto HPLC.
