Zusammenfassung
Aus Rindernetzhaut wurde ein Gangliosidpräparat mit einem Neuraminsauregehalt von 29,6 % erhalten (berechnet als N-Acetyl-neuraminsäure); der Phosphorgehalt lag unter 0,01 % ; der Hexosamingehalt betrug 5,53% (berechnet als N-Acetyl-D-galaktosamin).
Zur Gewinnung hochgereinigter Ganglioside wurde jetzt vorwiegend folgendes Verfahren angewendet. Fällung der homogenisierten Netzhant mit kaltem Aceton; Extraktion des Niederschlags mit einem Gemisch von Chloroform und Methanol (1:3); Vakuumtrocknung; wiederholte Lipoidtrennung nachFolch; Dialyse; Entsalzen mit Ionenaustauscher Dowex W-X 8; Entfernung der phosphorhaltigen Verbindungen mit der Aluminiumoxydsäule nachKlenk. — Durch Auskochen des getrockneten Chloroform-Methanol-Extraktes mit Aceton ließ rich (auch im SoxhletGerät) nur ein geringfügiger Reinigungseffekt erzielen. Bei Verwendung von heißem Ather löste sick ein erheblicher Gangliosidanteil aus dem Rohextrakt, insbesondere nach dessen Vorbehandlung mit Aceton.
Mit Hilfe der Dtinuschicht-Chromatographie ließen rich ausgiebig dialysierte Gangliosidpräparate in sechs Unterfraktionen trennen.
Summary
Preparations of gangliosides were obtained from retinas of cattle containing 29,6% neuraminic acid (assumed in the form of N-acetylneuraminic acid) and 5,5 % hexosamine (assumed in the form of N-acetylD-galactosamine). The contents of phosphorus was less than 0,01%.
In most cases, the following method was employed in order to obtain pure gangliosides : precipitation of the homogenized retinas with cold acetone; extraction of the precipitate with a mixture of chloroform and methanol (1:3); desiccation in vacuum; repeated separation from other lipoids usingJ. Folchs method; dialysis; acidic cation exchange resinDowex 50 W-X 8 ; removal of the phosphoric compounds in an aluminiumoxide column as devised byE. Klenk. — Boiling of the dried chloroformmethanol extract (even in the Soxhlet-boiler) did not contribute particularly towards purification. Hot ether dissolved a noticable fraction of the gangliosides out of the extract, especially, after it was treated with acetone.
Thin layer chromatography permitted separation of six fractions from the thoroughly dialysed ganglioside preparations.
