Summary
We determined the levels of some metabolites in the cornea stroma, the conjunctiva, the vitreous body, the aqueous humor and in the blood of rabbits. The glucose concentrations decreased from plasma to the aqueous humour and from there to the cornea stroma on one side and to the vitreous on the other side. Vice versa, we found a decrease of the concentrations of lactate and pyruvate from the cornea stroma and from the vitreous body to the aqueous humour and to the blood plasma. In the conjunctiva, the glucose levels were much higher and the lactate and pyruvate levels were considerably lower than in the cornea. The differences between conjunctiva and cornea stroma were less with regard to the levels of the adenosine phosphate compounds (ATP, ADP, AMP) and glucose-6-phosphate. We found relatively low levels of the intracellular phosphate compounds in the conjunctiva as well as in the cornea stroma according to the small number of cells and the large extracellular space per tissue volume. When the denuded surface of the stroma was rinsed with 0.9% saline in vivo, considerable amounts of glucose were found in the rinsed fluid.
After rinsing, the glucose levels were lower in the cornea stroma and in the conjunctiva compared to the unrinsed tissue. In the discussion, we considered, whether the concentration gradients may have some significance to the movement of metabolites in the avascular parts of the eye.
Zusammenfassung
Im Corneaparenchym, in der Conjunctiva, im Glaskörper und im Kammerwasser sowie im Blut von Kaninchen bestimmten wir die Spiegel verschiedener Metabolite. Die Glucose zeigte abnehmende Konzentrationen vom Plasma zum Kammerwasser und von dort zur Hornhaut einerseits und zum Glaskörper andererseits. Ein entgegengesetztes Konzentrationsgefälle ermittelten wir für Lactat und Pyruvat von der Hornhaut bzw. dem Glaskörper zum Kammerwasser und weiterhin zum Blutplasma. In der Conjunctiva lagen die Glucosespiegel wesentlich höher und die Lactat- und Pyruvatspiegel erheblich niedriger als in der Cornea. Ein geringerer Unterschied zwischen Conjunctiva und Corneaparenchym bestand im Hinblick auf den Gehalt an Adenosinphosphatverbindungen (ATP, ADP, AMP) und an Glucose-6-Phosphat. Entsprechend der geringen Zahl von Zellen und dem hohen Anteil des extracellulären Raumes am Gewebsvolumen fanden wir in der Conjunctiva und im Corneaparenchym nur relativ niedrige Spiegel der intracellulären phosphorylierten Metabolite. Wenn wir die bloße Oberfläche des Corneaparenchyms in vivo mit 0.9% NaCl bespülten, erschienen beträchtliche Mengen an Glucose in der Spülflüssigkeit. Dementsprechend waren die Glucosespiegel im Corneaparenchym und in der Conjunctiva nach dem Abspülen im Vergleich zu ungespülten Gewebsproben herabgesetzt. Es wurde diskutiert, ob die Konzentrationsgradienten Rückschlüsse auf Metabolitbewegungen in den gefäßlosen Teilen des Auges erlauben.
