Summary
With reference to earlier automatic procedures quantitative determinations are possible in different compartments of a single eye by a micromethod, analysing amino acids by elution chromatography. A complete chromatographic analysis can be done with 0.1 g of vitreous body or 0.03 g of a queous humour from rabbit or human eyes. There are significant differences of the amino acid content between vitreous and aqueous humour analogue to makromethods in rabbits. Cysteic acid and taurine in aqueous humour, glyeine, alanine and leucine in vitreous, aspartic acid, glutamic acid and phenylalanine in aqueous humour as well as in vitreous body show significant differences of concentration before and after hydrolysis of the perchloric extracts.
Zusammenfassung
In Anlehnung an ältere automatisierte Verfahren können durch eine Mikromethode der elutionschromatographischen AS-Trennung in verschiedenen Gewebsteileneines Auges Messungen vorgenommen werden. Es reichen dazu 0,1 g Glaskörper bzw. 0,03 g Vorderkammerpunktat beim Kaninchen für ein Chromatogramm aus, das natürlich in analoger Weise auch beim Menschenauge geschrieben werden kann. Die statistische Auswertung der Befunde an Kaninchenaugen zeigt signifikante Unterschiede des AS-Gehaltes zwischen Kammerwasser und Glaskörper in Übereinstimmung mit Makromethoden. Nach Hydrolyse des mit 0,5 n HClO4 enteiweißten Substrates treten sowohl im Kammerwasser als auch im Glaskörper signifikante Unterschiede für Asparaginsäure, Glutaminsäure und Phenylalanin auf, im Kammerwasser zusätzlich für Cysteinsäure und Taurin, im Glaskörper für Glycin, Alanin und Leucin.
