Summary
Potato cell walls were boiled at pH 6.1 to study the solubilization of pectic substances. It was shown that pectin was degraded by a β-elimination reaction. Ions accelerated this breakdown. partly by a specific effect. Calcium, cupric and ferrous ions retarded the solubilization of pectin in comparison with potassium. With calcium ions optimum stability against solubilization of pectin could be obtained when similar equivalents of both calcium and pectin acid groups were present. Calcium-binding anions—citrate, phytate and malate—favoured the solubilization of pectin.
Conversion of esterified into free acid groups in the pectin structure increased the binding activity towards calcium, so that less pectin became soluble. It was concluded that calcium was complexed more strongly by pectin than by potato starch.
Zusammenfassung
Der Verlust der interzellularen Kohäsion, ein Hauptmerkmal der Textur gekochter Kartoffeln, ergibt sich aus dem Verlust der verbindenden Eigenschaften des interzellular kohäsiven Materials, d.h. der Pektinsubstanzen. In einem Modellversuch wurde der Einfluss von Kartoffelbestandteilen auf die Löslichkeit von Pektin durch Kochen von isolierten Zellwänden mit verschiedenen Ionen und Stärke untersucht.
Der Mechanismus des Pektinabbaus bei pH 6,1 wurde als eine β-Elimination angesehen, was durch ein typisches Perjodat-TBS-Spektrum (Abb. 1) und durch eine erhöhte Reaktionsgeschwindigkeit bei erhöhtem pH (Abb. 2), die sich in löslicherem Pektin nach dem Kochen auswirkte, bestätigt wurde.
Calcium-, Kupfer- und Eisenionen verzögerten, im Vergleich zu Kalium, die Löslichkeit von Pektin (Abb. 3). Magnesium verzögerte die Löslichkeit nicht. Calcium jedoch erhöhte, verglichen mit Kalium, auch die Reaktionsgeschwindigkeit der β-Elimination (Abb. 4). Diese gegensätzlichen Wirkungen von Calcium zeigen die Fähigkeit dieses Ions, das Pektin in der Kartoffelzellwand in einem unlöslichen Zustand zu halten.
Optimale Stabilität gegen die Auflösung von Pektin wurde bei einem Verhältnis von Calcium zu Pektinsäureäquivalenten von 1–2 erzielt. Zugabe von Calciumbindenden Anionen, zum Beispiel Zitrat, Phytat und Malat, erhöhte die Löslichkeit des Pektins aus der Zellwand (Abb. 5). Sowohl Kationen wie Anionen beschleunigten den β-eliminierenden Abbau, zum Teil durch eine unspezifische Wirkung der Ionenstärke. Bei Erhöhung der Konzentration des Puffers erhöhte sich denn auch die Löslichkeit des Pektins während des Kochens (Abb. 6).
Fast vollständige Verseifung des zu 58% veresterten Zellwandpektins durch eine Pektinesterase (PE) erhöhte die Affinität für Calciumionen. In Anwesenheit von Calcium wurde weniger Pektin löslich, während die Calcium-bindende Fähigkeit der organischen Anionen ebenfalls abnahm (Tabelle 1).
Kartoffelstärke ohne gebundenes' Calcium übte keinen Einfluss auf die Pektinlöslichkeit aus (Abb. 7). Wenn Calciumionen in der Stärke vorhanden waren, wurden sie als teilweise in die Zellwände transferriert angesehen, was durch eine Abnahme der Löslichkeit von Pektin belegt wurde.
Résumé
La perte de la cohésion cellulaire, caractéristique principale de la texture de pommes de terre cuites, résulte de la perte des propriétés du ciment responsable de l'adhérence intercellulaire (substances pectiques). Dans cette étude, l'influence des constituants du tubercule sur la solubilité des pectines a été observée sur des membranes cellulaires isolées, pendant la cuisson, en présence de divers ions et d'amidon.
Le mécanisme de la dégradation des pectines à pH 6.1 a été assimilé à une réaction de β-élimination, ce qui est confirmé par le spectre typique du test périodate-TBA (Fig. 1) et par l'augmentation du taux de dégradation en fonction de l'élévation du pH (Fig. 2), conduisant à une teneur plus importante en pectines solubles après la cuisson.
Les ions calcium, cuivre et fer, diminuent la solubilité des pectines par rapport au potassium (Fig. 3). Le magnésium n'a pas cette action.
Le calcium, cependant, a aussi augmenté le taux de β-élimination en comparaison avec le potassium (Fig. 4). Ces effects opposés mettent en évidence l'aptitude de cet ion à maintenir les pectines dans un état insoluble au niveau de la membrane cellulaire.
L'optimum de stabilité des pectines a été obtenu avec un rapport calcium-pectines acides de 1–2. L'addition d'anions organiques séquestrant le calcium, tels le citrate, le phytate et le malate, augmente la solubilité des pectines de la membrane cellulaire (Fig. 5). Cations et anions ont accéléré la réaction de β-élimination, en partie par un effet non spécifique de force ionique. L'élévation de la molarité des solutionstampons augmente la solubilité des pectines durant la cuisson (Fig. 6).
La saponification presque totale des 58% de pectines estérifiées, contenues dans la membrane cellulaire, par une pectinestérase, augmente leur affinité pour le calcium. En présence de cet ion les pectines deviennent moins solubles tandis que l'effet séquestrant des anions organiques, vis-à-vis du calcium, diminue également (Tableau 1).
L'amidon libre de calcium n'a pas d'effet sur la solubilité des pectines (Fig. 7). Quand l'ion calcium est lié à l'amidon il peut être partiellement transféré à la membrane cellulaire et provoquer une diminution de la solubilité des pectines.
