Zusammenfassung
Eine nachhaltige Bekämpfung der durchFusarium solani f. sp.eumartii verursachten Verluste im Kartoffelbau könnte mit Hilfe der Resistenzzüchtung erzielt werden. Auf der Suche nach Resistenzgenen ist es von grossem Interesse, das genetische Potential auch der knollentragenden Kartoffelwildarten, welches in mehreren Genbanken zur Verfügung steht, zu evaluieren. Dieses setzt eine geeignete Testmethode voraus.
In der vorliegenden Arbeit wurden Kartoffelsämlinge von verschiedenenSolanum-tuberosum-Kreuzungen und knollentragenden Wildarten mit verschiedenen Methoden inokuliert. Es wurde festgestellt, dass sich die sogenannte ‘Agar’-Methode am besten zur Inokulation der Sämlinge eignet, da diese Methode schnell und in hohem Ausmass infiziert, ihre Handhabung einfach ist und Kontaminationen gut kontrolliert werden können. Ausserdem konnte mit der genannten Methode ein unterschiedliches Verhalten der Sämlinge verschiedenerSolanum-Arten gegenüber der Infektion mitF. solani f.sp.eumartii erkannt werden.
Summary
Lasting control of losses in the potato crop caused byFusarium eumartii can be achieved by breeding for resistance. In the search for resistant genes it is of particular interest to evaluate the genetic potential of tuber-bearing wild species available from several gene banks. This requires a suitable method of testing.
In the work described, various methods of inoculating potato seedlings of differentSolanum tuberosum crosses (Tables 1–3) and tuber-bearing wild species (Table 4) were tested. A comparison was made of the so-called ‘Agar’ method (cultivation of the fungus on 2% potato dextrose agar in Petri dishes, after three weeks sprinkling the uniform growth cover on the medium with a measured amount of sterilised, sieved soil, incubating one week at 20–25°C in closed Petri dishes and, finally, planting with seedlings) with the remainder of the published methods acceptable (Tables 1–3). The ‘Agar’ method proved the best suited to seedling inoculation as it yielded both a rapid and a high level of infection, manipulation is simple and contamination can be easily controlled. Furthermore it enabled different reactions of the seedlings of the variousSolanum spp. to infection byF. eumartii to be determined.
Variation of daylength to which the seedlings were exposed (length of period of illumination, change from light to darkness at intervals of 12 hours) after inoculation showed no clear indication of influence on infection (Table 5).
During the testing of the various methods of inoculation, repeated variation were observed in both the growth and the morphology ofF. eumartii conidia during the spread of the fungus from the initial culture (e.g. potato dextrose agar, wheat grains) into the sterilised soil. Particularly striking was the variation in the macroconidia (Fig. 4). The fungus sporulated very rapidly in the sterilised soil and formed typicalF. eumartii macroconidia, which probably influenced its pathogenicity.
Résumé
On peut réaliser une lutte efficace contre les pertes causées dans la culture de pomme de terre parFusarium eumartii par la recherche de la résistance. Pour trouver les gènes de résistance, il est particulièrement intéressant d'évaluer également le potentiel génétique des espèces sauvages tubérifères dont nous disposons dans de nombreuses banques de gènes. Ceci suppose une méthode de test adéquate.
Dans un travail précédent, diverses méthodes d'inoculation furent testées sur des plantules de pomme de terre de plusieurs croisementsSolanum tuberosum (tabl. 1 à 3) et d'espèces sauvages tubérifères (tabl. 4). On a comparé la méthode dénommée ‘méthode Agar’ (culture du champignon sur Agar-glucose de pomme de terre à 2% en boite de Pétri; après 3 semaines de multiplication, une quantité déterminée de terre tamisée stérilisée est épandue régulièrement sur le bouillon de culture en boites de Pétri fermées et conservées 1 semaine à 20–25°C; on transplante alors les plantules) avec les autres méthodes de test acceptables (tabl. 1 à 3); ainsi cette méthode est appliquée au mieux pour l'inoculation des plantules, car elle infecte rapidement et dans une mesure élevée, sa manipulation est simple et les contaminations peuvent être bien contrôlées. Au surplus on pouvait trouver, avec la méthode prénommée, un comportement variable des plantules des différentes espèces deSolanum (tabl. 4) à l'infection avecF. eumartii.
Le changement dans la durée d'éclairement des plantules (durée d'éclairement, alternance de périodes de 12 heures d'éc lairement et d'obscurité) après l'inoculation ne montrait aucune influence significative sur le résultat de l'infection (table 5).
Au cours du test des différentes méthodes d'inoculation, on a observé des changements répétés dans la croissance et dans la morphologie des conidies deF. eumartii, lesquels changements au cours du développement interne du champignon apparaissaient sur la culture finale (par ex. Agar-glucose de pomme de terre, grains de froment) dans la terre stérilisée. Particulièrement frappante était la variation des macroconidies (fig. 4). Le champignon sporulait très vite dans la terre stérilisée et produisait des macroconidies typiques deF. eumartii, grâce à quoi la pathogénéité était apparement influencée.
