Zusammenfassung
Es wird ein Überblick über Arbeiten zur Identifizierung von Kartoffelsorten aus Knollensaft gegeben, die auf der Proteinverteilung nach Elektrophorese basieren. Auf eine einfache plattenelektrophoretische Schnellmethode mit Auswertung anhand der Tafeln eines 1976 erschienenen ‘Index Europäischer Kartoffelsorten’ wird hingewiesen. Die an gleicher Stelle erfolgte numerische Wiedergabe von Eigenschaften und Anfälligkeiten der Sorten wird im Zusammenhang mit dem Fernziel einer rascheren Erzielung von Zuchterfolgen mittels ‘biochemischer Marker’ diskutiert.
Summary
Work with potato tuber proteins is reviewed in so far as it has a bearing on the identification of cultivars by their protein patterns after electrophoresis. The use of band positions as gene markers is seen as a possible further development in genetic evaluation. Numerical data on disease susceptibility and tables of genetical information contained in the ‘Index of European potato varieties’ are referred to as well as the electrophoretic spectra of the various cultivars. Problems of numerical assessment are discussed.
The various procedures in electrophoresis are critically evaluated and for reasons of simplicity and rapidity the use of flat plates of polyacrilamide gel in conjunction with photographic recording is recommended, using as experimental material the crude sap expressed from frozen tubers without further purification. Screening of densitograms did not give nearly as good results.
To facilitate identification of varieties, cv. Maritta was chosen for an internal standard, the main protein bands forming the basis for 9 groups. The position of the less pronounced bands and the esterase pattern serve for further characterisation. A photograph of typical protein and esterase patterns is given (Fig. 1).
The publication of the ‘Index’ should promote future international cooperation and lead to standardisation of the tests.
Résumé
Le travail débute par une étude des travaux publiés sur les protéines de la pomme de terre et utilisés pour l'identification des variétés à l'aide de leurs spectres de protéines obtenus par l'électrophorèse. L'utilisation des positions des bandes comme traceur génétique semble présenter une possibilité pour améliorer la taxation génétique. Il est rappelé que l'‘Indexe européen des variétés de pommes de terre’ contient, en plus des spectres d'électrophorèse, des données numériques sur la sensibilité et l'origine des variétés. Les problèmes en relation avec la taxation numérique sont discutés.
Les différents procédés d'électrophorèse sont examinés et il est recommandé comme méthode rapide à pouvoir séparateur élevé l'électrophorèse sur plaques de gel de polyacrylamide et leur interprétation par photographie. Le screening des densitogrammes ne donne pas encore d'information aussi valables. Les échantillons se composent de jus cru et centrifugés (sans purification) de pommes de terre congelées. L'obtention Maritta a été utilisée comme standard interne pour faciliter l'identification des variétés. Les zones principales de protéines présentent la base pour 9 groupes (fig. 1). Les positions des zones moins marquées et les échantillons d'estérase sont utilisés pour une description complémentaire. La figure présente des échantillons caractéristiques de protéines et d'estérases. Le but de cet ‘indexe’ est de favoriser la collaboration internationale et la standardisation des tests.
