Abstract
Endorphins are endogenous substances which mimic the action of morphine. The existence of specific binding sites for morphine-like drugs (opiate receptors) in the brain and in peripheral tissues led to the discovery of the endorphins. All hitherto isolated endorphins are structurally related to the pituitary hormoneΒ-lipotropin. Hormones of the pituitary gland which appear to be present in the brain albeit in much smaller quantities, are involved in brain functions. The influence of these hormones is localised in certain fragments, designated as neuropeptides, which themselves are devoid of the classical endocrine action of their, precursor molecules.
In addition to their opiate-like activity, the endorphins appear to modulate adaptive behaviour. In particular the neuropeptide des-tyrosine-γ-endorphin (Dt γ e) produces effects on animal behaviour which are in some respects comparable to those of neuroleptic drugs. A clinical study with schizophrenic patients showed anti-psychotic activity ofDt γ e. These observations suggest that a decreased availability ofDt γ e due to an ‘inborn error’ in the metabolism of endorphins may be an aetiological factor in schizophrenic psychosis.
Samenvatting
Endorfinen zijn endogene Stoffen die de werking van morfine nabootsen. De aanwezigheid van specifieke bindingsplaatsen voor morfine-achtige Stoffen (opiaatreceptoren) in de hersenen heeft bijgedragen tot de ontdekking van deze substanties. De tot nu toe geÏsoleerde en gekarakteriseerde endorfinen zijn verwant aan het hypofysehormoonΒ-lipotropine. De hormonen van de hypofyse die, hoewel in kleinere hoeveelheden, ook in de hersenen aanwezig zijn, reguleren niet alleen de activiteit van perifeer gelegen endocriene organen, maar ook verschillende functies van het centrale zenuwstelsel. Voor hun invloed op hersenfuncties is echter niet het gehele molekuul nodig, wat voor hun klassieke endocriene werkzaamheid wel het geval is. Bepaalde brokstukken, neuropeptiden genaamd, blijken dezelfde effecten te bewerkstelligen als het moederhormoon. Deze neuropeptiden zijn vooral betrokken bij het aanleren en handhaven van nieuwe gedragspatronen.
Vastgesteld is dat de endorfinen, behalve hun opiaatachtige werking, ook een belangrijke regulerende invloed uitoefenen op adaptief gedrag. Uit voortgezet onderzoek is gebleken dat de invloed op het gedrag van proefdieren van met name het peptide destyrosine-γ-endorfine (Dt γ e) in sommige opzichten vergelijkbaar is met dat van bekende neuroleptica. In een klinisch onderzoek bij schizofrene patiënten werd een duidelijk antipsychotische werking vanDt γ e geconstateerd. Deze waarnemingen doen vermoeden dat de schizofrene psychose een gevolg is van een tekort aan het neuropeptideDtγe veroorzaakt door een ‘inborn error’ in het metabolisme van de endorfinen.
