Summary
The possibilities of a high-speed liquid chromatographic (HPLC) method using ion exchange and reversed-phase techniques for dihydroxyphenylalanine (dopa) and the catecholamines dopamine, noradrenaline, and adrenaline in combination with a reaction detector are described. The reaction detector comprises a fluorimeter and a reaction unit with liquid dose-pump. In the reaction unit the substances coming from the HPLC column react with ethylenediamine (AeDA) to form fluorescent products. By combining the HPLC separation with the selective AeDA-reaction and fluorimetric measurement a separate and sensitive analysis of components using isopropyl noradrenaline as internal standard is possible in about 20 minutes.
Zusammenfassung
Es werden die Möglichkeiten einer hochdruck-flüssigkeitschromatographischen (HPLC-) Analyse durch lonenaustausch und „reversed-phase”-Chromatographie für Dihydroxyphenylalanin (Dopa) und die Katecholamine Dipamin, Noradrenalin und Adrenalin in Verbindung mit einem Reaktionsdetektor beschrieben. Der Reaktionsdetektor besteht aus einem Fluorimeter und einer Reaktionseinheit mit Flüssigkeits-Dosierpumpe. In der Reaktionseinheit findet die Umsetzung der von der HPLC-Säule kommenden Substanze mit Äthylendiamin (AeDA) zu fluoreszierenden Reaktionsprodukten statt. Durch die Verbindung der HPLC-Trennung mit der selektiven AeDA-Reaktion und fluorimetrischen Messung ist eine getrennte und empfindliche Analyse der Substanzen unter Verwendung von Isopropyl-noradrenalin als inneren Standard in etwa 20 Minuten möglich.
