Summary
In the course of our studies on the chromatographic and electrophoretic mobility of α-amylase from Bacillus subtilis we have shown that the changes in mobility of this enzyme in relation to the pH of the medium 1) are the result of a reversible transformation between monomeric and dimeric form; 2) cannot be explained except by the participation of zinc in this transformation.
By means of the results obtained with these two techniques, we have succeeded in characterizing the monomeric and dimeric forms of this α-amylase and in investigating some of its properties.
By zone electrophoresis in gelled medium we have determined: 1) the apparent isoelectric pH values of the monomeric and of the dimeric form of α-amylase from B. subtilis; 2) the regions of pH values where one of the two forms is dominating (pH 5–6 for the monomeric form and pH 8–10 for the dimeric form), as well as the pH region where the transformation between monomeric and dimeric form can take place (pH 6–8). By this technique it can also be shown that the monomeric-dimeric transformation takes place together with a change in the net electric charge of the molecule. This change in the net charge apparently cannot be explained except by the formation of an additional negative ion during the fixation of zinc for the formation of the dimeric form of the enzyme, this negative ion perhaps results from the ionization of a peptide bond at a point of the enzyme molecule which evidently must be well defined.
By column chromatography on grains of cross-linked dextrane gel we have shown that the variations in mobility of the zone of α-amylase in relation to the pH of the medium can only be explained by the existence of the reversible equilibrium:
Taken together, the chromatographic and electrophoretic results warrant the conclusion that the monomeric form of α-amylase has a positive net charge, while the net charge of the dimeric form is negative in those pH regions where each of these forms exists. Finally, these results help to explain the aspect of the electrograms in gelled medium such as they were obtained for α-amylase of B. subtilis.
Résumé
Au cours d'études sur les mobilités chromatographiques et électrophorétiques de l'α-amylase deBacillus subtilis, nous avons montré que les changements de mobilité de cet enzyme en fonction du pH du milieu: 1°) résultent d'une transformation réversible monomèredimère, 2°) ne peuvent s'expliquer que par la participation du zinc à cette transformation.
Grâce aux résultats obtenus par ces deux techniques expérimentales, on a pu caractériser les formes monomère et dimère de cette α-amylase et étudier quelques-unes de leurs propriétés.
Par électrophorèse de zones en milieu gélifié, on a pu déterminer: 1°) les pH isoélectriques apparents du monomère et du dimère de l'α-amylase deB. subtilis, 2°) les domaines de valeurs de pH où l'une des deux formes domine (pH 5–6 pour le monomère, pH 8–10 pour le dimère) ainsi que le domaine de pH où la transformation monomère-dimère peut avoir lieu (pH 6–8). Par cette technique on a pu, également, montrer que la transformation monomère-dimère a lieu avec changement de la charge nette de la molécule. Ce changement de la charge nette ne semble pouvoir s'expliquer que par l'apparition d'un ion négatif supplémentaire lors de la fixation du zinc pour la formation du dimère de l'enzyme, ion négatif résultant peutêtre de l'ionisation d'une liaison peptidique en un point de la molécule d'enzyme qui ne peut, d'une manière évidente, qu'être bien déterminé.
Par chromatographie sur colonne de grains de gel de dextrane réticulé, on a montré que la variation de mobilité de la zone d'α-amylase en fonction du pH du milieu ne peut s'expliquer que par l'existence de l'équilibre reversible:
L'ensemble des résultats chromatographiques et électrophorétiques permet d'affirmer que le monomère de l'α-amylase a une charge nette positive tandis que le dimère a une charge nette négative dans les domaines de pH où chacune de ces formes existe. Enfin, ces résultats permettent d'expliquer l'aspect des électrogrammes en milieu gélifié obtenus pour l'α-amylase deB. subtilis.
