Summary

An immunofluorescent method for the diagnosis of African swine fever (ASF) is described. Diagnostic samples inoculated onto PK15 swine kidney cell line monolayers on coverslips, are acetone-fixed and stained with fluorescein isothiocyanate-conjugated ASF convalescent serum globulin at 24, 48 and 72 hours after inoculation. Specific fluorescence is detected in ASF infected cells as granular or globular inclusions. The method detected ASF virus in materials from some infected pigs which produced no haemadsorption at all, or only after two to three serial passages in swine buffy coat cultures. This test may provide a sensitive and rapid method for diagnosing ASF in chronic infection.

Résumé

On décrit ici une méthode d'immunofluorescence pour le diagnostic de la peste porcine africaine (A.S.F.). Des prélèvements reçus pour diagnostic ont été inoculés à des cultures sur lamelle de la lignée cellulaire de rein de porc PK15; ces lamellas ont été colorées, 24, 48 et 72 heures après l'inoculation, par des globulines de porc convalescent d'A.S.F. conjuguées à l'isothiocyanate de fluorescéine, après fixation à l'acétone. La fluorescence spécifique fut décelée dans les cellules infectées sous forme d'inclusions granulaires ou globulaires.

Cette méthode a décelé le virus de la peste porcine africaine dans du matériel prélevé sur des porcs infectés et qui ne produisait pas du tout d'hémadsorption, ou seulement après deux ou trois passages en série sur culture de leucocytes de porc. Ce test peut donc constituer une méthode rapide et sensible pour le diagnostic des formes chroniques d'A.S.F.

Se describe un método de inmunofluorescencia para el diagnóstico de la fiebre Africana porcina (ASF). Las muestras diagnósticas fueron inoculadas en monocapas de la linea celular PKII de riñón porcino cultivadas en laminillas de vidrio. Estas laminillas fueron fijadas en acetona y coloreadas con fluoresceina de isothiociantato conjugado a globulinas de suero convaleciente de ASF a las 24, 48 y 72 horas después de la inoculación. Fluorescen cia específica en la forma de inclusiones granulares ó globulares fueddetectada en células infectadas con ASF.

El método detectó el virus de ASF en materiales de algunos porcinos infectados el cual no producia hemadsorbcion ó solo despunes de 2 a 3 pases seriados en cultivos de “buffy coat” de cerdos. Esta prueba puede ser un método rápido y sensitivo para el diagnóstico de ASF en infecciones crónicas.
