Zusammenfassung
Untersucht wurden hypoplastische Defckte der Schmelzoberflächen stark fluorotischer permanenter Unterkiefer-Backenzähne des Rehs. Die Form der Hypoplasien variierte zwischen ausgedehnten, relativ flachen Einbuchtungen der Oberfläche und eng begrenzten, trichterartig in den Schmelzmantel hineinreichenden Vertiefungen. Der Übergangsbereich zwischen den Hypoplasien und den angrenzenden Bereichen normaler Schmelzmächtigkeit wies jeweils ein konvex gerundetes Profil auf. Untersuchungen von Dünnschliffen durch die Hypoplasien im normalen und polarisierten Licht zeigten, daß die in diesen Bereichen dichter gescharten Retziuslinien sehr genau die Konturen der Oberflächendefekte nachzeichneten. Die Betrachtung im Rasterelektronenmikroskop ergab, daß die Wandzonen der Hypoplasien durch zahlreiche Ameloblastenfortsatzgrübchen strukturiert waren, die sich auch in den Übergangsbereichen zum angrenzenden Schmelz und am Boden der flachen Defektzonen fanden. Aus diesen, sowie den Beobachtungen einer vorhergehenden Studie (Kierdorf et al. 1993) wird gefolgert, daß die Entstehungsursache der Hypoplasien eine mehr oder weniger starke Einschränkung der sekretorischen Aktivität der Ameloblasten war. Eine Ablösung der Ameloblasten von der Schmelzbildungsfront und die Ausbildung subameloblastischer Cysten, wie sie nach Injektion hoher Fluoriddosen bei Nagern beobachtet wurden, ist dagegen als Entstehungsmodus der hier beschriebenen hypoplastischen Schmelzdefekte an den Rehzähnen auszuschließen.
Summary
Hypoplastic enamel surface lesions were studied in severely fluorosed permanent mandibular cheek teeth. The shape of the hypoplasias varied between large, rather shallow depressions of the enamel surface and narrow lesions running deep into the enamel. The areas of transition between hypoplasias and surrounding enamel were of a convexly rounded outline. Studies of thin ground sections through hypoplasias viewed in normal and polarized light showed the (crowded) Retzius lines to exactly follow the shape of the hypoplastic surface lesions. As was revealed by scanning electron microscopy, the bottom of the shallow defects as well as the walls of the lesions and the transitional area between hypoplasias and surrounding surface enamel were covered by Tomes' process pits. Based on the present findings and that of a previous study (Kierdorf et al. 1993), we interprete these enamel surface hypoplasias to be the result of a more or less intense reduction in the secretory activity of the ameloblasts. On the basis of our observations, detachment of the ameloblasts from the forming enamel surface and formation of subameloblastic cysts, that were observed in rodents after injection of high doses of fluoride, can be excluded as the way by which formation of the hypoplasias described in this study had occurred.
Résumé
On a analysé les déficiences hypoplastiques causées aux surfaces d'émail de molaires définitives, fortement fluorées, du maxillaire inférieur du Chevreuil. Les hypoplasies se présentaient sous une forme variable, allant d'anses étendues et relativement plates en surface jusqu'à des cavités ressemblant à des entonnoirs, bien délimitées se prolongeant dans le manteau d'émail. La zone de transition entre les hypoplasies et les régions avoisinantes présentant une couche d'émail normale mais abondante révèle chaque fois un profil arrondi et convexe. Des examens de coupes minces à travers les hypoplasies en lumière normale et polarisée montrent qu'au niveau de ces zones, les lignes de Retzius, telles des rayures très denses, suivent très précisément les contours des défauts de surface. L'examen par microscopie électronique à balayage révéla que les zones pariétales des hypoplasies étaient structurées par de nombreux appendices sous forme de fossettes d'améloblastes; celles-ci s'observaient également dans les zones de transition à proximité de l'émail et au fond des zones planes de déficience. On déduit de ces observations ainsi que de celles d'une étude antérieure (Kierdorf et al. 1993) que les causes d'apparition des hypoplasies résident dans une activité plus ou moins réduite des améloblastes. Une dissolution des améloblastes par le front de formation d'émail et la formation de cystes subaméloblastiques tels qu'on les observe après injection de doses élevées de Fluoride chez les Rongeurs, est par contre à rejeter comme initiateur des déficiences d'émail hypoplastiques décrites ici à propos des dents du Chevreuil.
