Summary
In the technological processing of potatoes undesirable changes of taste are often caused by unsaturated fatty acids. Polar lipids are usually more unsaturated as neutral fats from the same tissue. The isolation of these compounds and their subsequent characterization was carried out by the development of a separation process.
After purification with salt water solutions and column-type chromatography seperation of other substances as well on Sephadex G 25 as on silicic acid, a lipid extract derived from vacuum freeze dried raw potatoes led on polar lipids.
The fractionating on DEAE-cellulose and the separation of the individual fractions was carried out by partition chromatography and gel chromatography.
The compounds isolated by the separation process were classified in the following manner on the basis of their properties on the different column types, by thin layer chromatography and by chemical analysis of their hydrolysis products: sterol-glycerol-ester, sterol-glycerol, monogalactosyl-diglyceride, digalactosyl-diglyceride, three cerebrosides, phosphatidyl-choline, phosphatidyl-ethanolamine, three phosphatidyc acids, phosphatidyl-serine, phosphatidyl-inositol, and a sulfolipid.
The distribution of the polar lipid class was established by gravimetrical estimation of the separated fractions and the fatty acids of the separated compounds, after transfer to the methyl esters, by gaschromatography.
Zusammenfassung
Bei der technologischen Kartoffelverarbeitung werden bisweilen uner-wünschte Geschmacksabwandlungen durch oxydativen Abbau ungesättigter Fettsäuren hervorgerufen. Polare Lipoide sind meist ungesättigter als Neutralfette aus dem gleichen Gewebe. Die Isolierung dieser Verbindungen und ihre anschließende Charakterisierung erfolgte über einen geeigneten Trennungsgang.
Ein aus gefriergetrockneten Rohkartoffeln extrahierter Lipoidextrakt führte nach Reinigung mit wäßrigen Salzlösungen und säulenchromatographischer Abtrennung weiterer Substanzen an Sephadex G 25 sowie Kieselsäure zu den polaren Lipoiden. Die Fraktionierung an DEAE-Cellulose und die Trennung der einzelnen Fraktionen geschah mittels Verteilungs- und Gelchromatographie. Durch den Trennungsgang isolierte Verbindungen wurden auf Grund ihrer Eigenschaften auf den einzelnen Säulentypen, durch Dünnschichtchromatographie und durch Untersuchung ihrer Hydrolysenprodukte wie folgt charakterisiert: Steringlykosid-ester, Steringlykosid, Monogalaktosyl-diglycerid, Digalaktosyl-diglycerid, drei Cerebroside, Phosphatidyl-cholin, Phosphatidyl-äthanolamin, drei Phosphatidsäuren, Phosphatidyl-serin, Phosphatidyl-inosit und ein Sulfolipid.
Die Verteilung der polaren Lipoidklassen wurde durch Gravimetrie der getrennten Fraktionen ermittelt und die Fettsäuren der getrennten Verbindungen, nach Überführung in die Methylester, gaschromatographisch bestimmt.
