Abstract
Esterase isozymes were studied in bone extracts from various species and in bone from newborn rabbits. Bone was frozen in liquid nitrogen and crushed. The isozymes were extracted with isotonic saline and separated by electrophoresis on starch gel. The isozyme pattern differed between species and between various developmental stages in the same species. Using specific enzyme inhibitors it was found that the esterase fractions identified in bone were presumably nonspecific organophosphate-sensitive esterases.
Résumé
Les isozymes d'estérase sont étudiées dans des extraits d'os d'espèces variées et dans l'os de lapin nouveau-né. L'os est congelé dans l'azote liquide puis pulvérisé. Les isozymes sont extraites dans du sérum physiologique isotonique et séparées par electrophorèse sur gel d'amidon. La distribution en isozyme diffère d'une espèce à l'autre et à divers stades de développement, dans la même espèce. En utilisant des inhibiteurs d'enzymes spécifiques, il semble que les fractions d'estérase identifiées dans l'os soient vraisemblablement des estérases nonspécifiques, sensibles à l'organophosphate.
Zusammenfassung
Esterase-Isoenzyme wurden in Knochenextrakten verschiedener Species und in Knochen neugeborener Kaninchen untersucht. Die Knochen wurden in flüssigem Stickstoff tiefgefroren und anschließend in einem kalten Stahlmörser zerrieben. Die Isoenzyme wurden dann mit isotonischer Kochsalzlösung extrahiert und mittels Stärkegelenkelektrophorese getrennt. Die Isoenzymmuster unterschieden sich zwischen den verschiedenen Species und auch zwischen den verschiedenen Entwicklungsstadien innerhalb derselben Species. Durch Anwendung von spezifischen Enzyminhibitoren konnte nachgewiesen werden, daß die im Knochen identifizierten Esterasefraktionen vermutlich unspezifische, auf organische Phosphate empfindliche Esterasen sind.
