In Affennieren-Zellkulturen, die mit dem Echo-Virus Typ 9 infiziert worden waren, gelang der Nachweis des Erregers mittels fluorescierender monotypischer Antikörper ausschließlich im Cytoplasma der Kulturzellen.

Der Nachweis von infektiösem Echo-Virus-Typ-9 in der Zelle geht parallel mit der morphologischen Erfaßbarkeit. Die Viruskonzentration war intracellulär anfangs (nach etwa 3 1/2 Std) höher als im Außenmedium (flüssige Phase).

Nach der Infektion wurden elektronenoptisch morphologische Veränderungen in der Zelle beobachtet. Neben Alterationen an den Mitochondrien entstanden blasige Gebilde im Cytoplasma; sie enthielten etwa 30 mμ große osmiophile Partikeln. Weiteren Untersuchungen muß es vorbehalten bleiben, diese Veränderungen zu differenzieren. Ein funktioneller Anteil des Ergastoplasma und der Mitochondrien wird bei der Virussynthese auf Grund ihres morphologischen Verhaltens diskutiert.

Als Ursache der beobachteten anfänglich schwachen, später jedoch erheblich stärkeren Fluorescenz infizierter Kulturzellen nach Beladung mit fluorescierenden Antikörpern wird eine Permeabilitäts-Vermehrung der Zellmembran im Verlaufe der ersten Stunden nach der Infektion erwogen. Die anfänglich geringe Fluorescenz könnte aber auch durch ein verzögertes Auftreten von spezifischem Virusprotein oder aber durch eine disseminierte Verteilung der Echo-Virus-Typ-9 Partikeln (zu geringes optisches Auflösungsvermögen) erklärt werden.
