A technique for controlled sensory denervation of the rabbit cornea

Abstract

Complications arising from the procedure to produce denervation of the cornea have made experiments on the trophic role of trigeminal nerve difficult. In this study, controlled lesions of the rabbit trigeminal ganglion were produced by radio-frequency thermocoagulation, delivered by a needle electrode inserted through the soft palate of the mouth. The advantages of the procedure were low mortality (<5%), permanent destruction of sensory neurons, and absence of spurious ocular effects. Two lesion sizes were reliably produced:

1. The smaller circumscribed lesion resulted in loss of corneal sensitivity, but preserved spontaneous blinking.

2. The larger lesion extended across the ganglion, abolished corneal sensation and spontaneous blink activity.

Zusammenfassung

Bisherige Methoden zur Denervierung der Kornea erschwerten Experimente über die trophische Rolle des N. trigeminus wegen häufiger Komplikationen. In der vorliegenden Studie werden erstmals kontrollierte Diathermiekoagulationen mit einer Nadelelectrode, die durch den weichen Gaumen des Kaninchens geführt wird, im Trigeminusganglion erzeugt. Die Vorteile dieser Technik sind eine niedrige Mortalitätsrate (<5%), permanente Destruktion sensorischer Neurone und Vermeidung falsch interpretierbarer klinischer Veränderungen des vorderen Segmentes. Zwei Typen von ganglionären Läsionen wurden zuverlässig erzeugt:

1. Eine umschriebene Läsion führte zum Verlust der Corneasensibilität, jedoch war die spontane Blinkaktivität erhalten.

2. Eine den ganzen Querschnitt des Trigeminusganglions umfassende Läsion hatte eine aufgehobene Corneasensibilität und Absenz der spontanen Blinkaktivität zur Folge.
