Summary
In addition to NaCl, KCl and glucose, calcium ions are necessary to support the viability of rabbit kidney cells. These substances are also sufficient for the formation of giant cells after infection with giant cell forming Herpes-virus-strains, not, however, for the synthesis of infectious viria. Relative lack of calcium can be compensated for by histidin or EDTA. Zinc ions block the formation of giant cells within the first 3 1/2 hours post infection, 2,4-Dinitrophenol, like Zn++, inhibits giant cell formation up to the same time, whereas phloridzin has no effect.
Zusammenfassung
Es wird über den Ionen- und Aminosäurebedarf der herpesinfizierten Zelle berichtet. Als Merkmal der Wirksamkeit prüften wir die Lebensfähigkeit der Zellen und die Befähigung zur Riesenzellbildung.
Die Lebensfähigkeit der Zelle ist außer von NaCl, KCl und Glucose (Isotonie) vor allem von [Ca++] abhängig. Diese Salze zusammen ermöglichen auch die Entstehung von Riesenzellen nach der Infektion mit riesenzellbildenden Varianten des Herpes-Virus hominis, nicht aber die Synthese infektiöser Viria. Ein relativer Ca++-Mangel kann durch Histidin kompensiert werden. Für diese Wirkung sind die freie NH2- und die Imidazolgruppe erforderlich. Die Histidin-Wirkung kann durch Chelatbildner, wie z. B. EDTA, nachgeahmt werden.
Von sechs zweiwertigen Metall-Ionen ist nur Zn++ in der Lage, die Riesenzellbildung durch Herpes-Virus hominis zu blockieren, wenn es innerhalb der ersten 3 1/2 Std gegeben wird. Die gegenseitige Abhängigkeit der Riesenzellbildung von [Ca++], [Zn++] und Histidin oder anderen Chelatbildnern legt die Annahme nahe, es bestehe ein leicht störbares Gleichgewicht zwischen Ca++ und Zn++. Dieses Gleichgewicht ist offenbar für die Lebensäußerung der Zelle nach Virusinfektion unerläßlich.
2,4-Dinitrophenol wirkt bis zum gleichen Zeitpunkt wie Zn++ hemmend auf die Riesenzellbildung ein, während Phloridzin ohne Einfluß ist.
