Zusammenfassung
Am isolierten Froschherzen wurden bei isotonischer und isometrischer Kontraktion das biphasische Eg und mit einer Saugelektrode das monophasische Aktionspotential registriert. Dabei konnte mit der früher beschriebenenQT-Verkürzung bei isometrischer Kontraktion eine Formänderung des MAP beobachtet werden. Bei isometrischer Fixierung des gefüllten Ventrikels kommt es zu einem Plateauverlust mit steilerem Verlauf und Verkürzung der Phase 2 des MAP. Außerdem wird im Verlauf der Phase 3 die schnelle Repolarisation unterbrochen und durch ein langsam dem Verletzungspotential zustrebendes negatives Nachpotential fortgesetzt. Die Veränderung treten mit der ersten isometrischen Kontraktion auf und sind mit der ersten isotonischen Kontraktion nicht mehr nachweisbar.
Während sonst ein Plateauverlust mit einer Abnahme der Kontraktion einhergeht, kommt es in diesen Versuchen bei zunehmender Füllung und der damit einhergehenden Zunahme des isometrisch entwickelten Druckanstiegs zu einem stärkeren Plateauverlust. Bei wenig gefülltem Ventrikel treten keine Veränderungen in der Form des MAP auf.
Diese Befunde werden als Zeichen dafür gewertet, daß außer der Koppelung von der Erregung zur Kontraktion auch eine Rückkoppelung von der Kontraktion zur Membran existiert.
Auf Grund der Befunde über die zeitlichen Beziehungen zwischen isometrischer und isotonischer Kontraktion des ganzen Herzens und dem MAP wird diskutiert, ob die theoretisch bei elektromechanischen Untersuchungen zu fordernden kleinsten Muskelfaserpräparate nicht sowiet in ihrer Kontraktilität geschädigt werden, daß an solchen Präparaten eine zeitliche Korrelation zwischen Kontraktion und Aktionspotential nicht mehr sicher möglich ist.
Summary
Monophasic action potentials from a suction electrode and unipolar electrograms were recorded from an isolated frog heart contracting under isotonic and isometric conditions. TheQT-shortening under isometric contractions, described earlier, was accompanied by alterations in the monophasic action potential. Isometric contraction of the filled ventricle results in a shortening and steepening of the slope of phase 2 of the action potential. Repolarisation in phase 3 is interrupted sharply after about one third of its course and continues slowly as a negative afterpotential, since theT-Wave corresponds to the sharp bend in phase 3. The changes appear with the first isometric and disappear with the first isotonic contraction. Increase of filling of the ventricle results in an increase of isometrically developed pressure and a decrease of the duration and slope of the plateau phase in contrast to the usual decrease of contractile force accompanying a shortened plateau. Contraction with little filling causes no change of the action potential.
These results are taken as a sign for the existence of a coupling mechanism in the direction of contraction to excitation and not only the evident excitation-contraction direction.
The results of correlations of isometric and isotonic contractions of the whole heart with the monophasic action potential raise the question, wether muscle fiber preparations, generally used for micropuncture, are impaired in their contractility so far that definite results about time relations between excitation and contraction may not be made, however, desirable such small preparations may be from a theoretical standpoint.
