Summary
The influence of various experimental conditions (time, temperature, red cell concentration, pH, ion content) on the haemagglutination and on the adsorption of haemagglutinins to blood cells was investigated for several viruses of group II (types 19 and 26, in some experiments for all other prototypes). Maximal adsorption occurred within 5 min. The extent of adsorption is dependent on the respective virus type and, in some instances, on the blood cell species (rat, man) and on temperature. At 45° C haemagglutination is absent for several types with human cells, for type 23 with rat cells. A partial elution may be obtained with types 19 and 26 and human blood cells by means of a temperature shift from 4 to 45° C. A treatment of human blood cells with influenza A 2 virus or with adenovirus 2 (application of the erythrocyte receptor-modifying factor,Kasel et al.) diminished their agglutinability for types 10, 13, 19, 26, 27, and 9–15, while they remained agglutinable for types 8 and 9. Rat blood cells treated with adenovirus 2 lost their agglutinability for types 23 and 29 only.
Zusammenfassung
Der Einfluß verschiedener Reaktionsbedingungen (Zeit, Temperatur, Erythrocyten-Konzentration, pH-Wert, Ionengehalt der Lösung) auf die Hämagglutination und auf die Adsorption der Hämagglutinine an die Blutzellen wird für verschiedene Typen der Gruppe II (Typ 19 und 26, zum Teil alle weiteren Prototypen) untersucht. Die Adsorption war bereits nach 5 min maximal. Sie ist in ihrem Ausmaß von dem Virustyp und in einigen Fällen von der Blutkörperchenart (Ratte, Mensch) und von der Temperatur abhängig. Bei 45° C fällt die Hämagglutination bei einigen Typen mit Menschen-, bei Typ 23 mit Rattenblutkörperchen negativ aus. Eine partielle Elution gelingt bei den Typen 19 und 26 von Menschenblutkörperchen durch Temperaturverschiebung von 4° auf 45° C. Eine Behandlung von Menschenblutkörperchen mit Influenza A 2-Virus bzw. mit Adenovirus 2 (Anwendung des die Receptoren der Erythrocyten modifizierenden Faktors,Kasel et al.) verminderte ihre Agglutinierbarkeit für die Typen 10, 13, 19, 26, 27 und 9–15, während sie für Typ 8 und 9 voll agglutinierbar blieben. Mit Adenovirus 2 behandelte Rattenblutkörperchen verloren ihre Agglutinierbarkeit lediglich für die Typen 23 und 29.
