Summary
The presence of neuraminidase activity in aMycoplasma gallisepticum strain was demonstrated by colorimetric determination of free N-acetylneuraminic acid released from various neuraminic acid containing substrates (N-acetylneuraminyllactose, human acid α1-glycoprotein, human transferrin and rabbit serum) by the test mycoplasma preparation. The enzyme activity was mostly cell-bound, activated by Ca.. ions (optimum Ca.. concentration: 0.0001 M) and inactivated by heat treatment (70°C for 30 min). The optimum pH for the low molecular weight substrate i.e. N-acetylneuraminyllactose was pH 4.5 and for the glycoproteins pH 5.8. The Michaelis constantK m as determined for N-acetylneuraminyllactose at pH 4.5 was 3.0×10−3 M, for human acid α1-glycoprotein at pH 5.8 was 5.0×10−4 M and for human transferrin at pH 5.8 was 3.5×10−4 M. The highest level of neuraminidase activity was obtained in 2–3 days old cultures, then the level started decreasing and in 5 days old cultures less than 10% of the maximum activity could be detected. The addition of catalase inhibited the fall of enzyme activity to some extent.
Zusammenfassung
Der Nachweis des Enzyms Neuraminidase beiMycoplasma gallisepticum wurde durch colorimetrische Bestimmung der aus verschiedenen Neuraminsäure-haltigen Substraten (N-Acetylneuraminyllactose, Human-saures α1-Glykoprotein, Human-Transferrin, Kaninchenserum) freigesetzten N-Acetylneuraminsäure geführt. Das größtenteils zellgebundene Enzym läßt sich durch Ca..-Ionen aktivieren (optimale Ca..-Ionenkonzentration: 0,0001 m) und durch Hitze inaktivieren (70°C, 30 min). Das pH-Optimum liegt für die niedermolekulare N-Acetylneuraminyllactose bei pH 4,5, für die Glykoproteine bei pH 5,8. Die Michaelis-KonstantenK m wurden bestimmt. Sie betragen für N-Acetylneuraminyllactose bei pH 4,5 3,0 · 10−3 m, für Hum.-saures α1-Glykoprotein bei pH 5,8 5,0 · 10−4 m und für Hum.-Transferrin bei pH 5,8 3,5 · 10−4 m. Die Neuraminidase-Aktivität erreicht bei 2–3 Tage alten Kulturen ihr Maximum, dann fällt sie ab und besitzt in 5 Tage alten Kulturen nur noch weniger als etwa 10% ihrer maximalen Aktivität. Der Zusatz von Katalase hemmt den Abfall der Enzymaktivität etwas.
