Summary
Following the principle of the immune-adherence-reaction we elaborated a method which allows the assay of virus-specific or cell-specific antigens upon or inside of membranes of living cells in vitro: Immune-Adherence-Hem-adsorption.
During the course of the Herpes virus hominis synthesis 4 hours p. i. virus-specific antigens appear in the membranes of rabbit kidney cells and other cell types. Application of cycloheximide (2 hours p. i.) prevents immune-adherence-hem-adsorption and similarly the CPE (cell rounding or giant cell formation). But not anymore does prevent application of cycloheximide at 31/2 hours p. i. both activities. The process of giant cell formation does not influence the intensity of this hemadsorption in respect of cell-specific Forssman antigens of guinea pig kidney cells in vitro. The immune-adherence-hemadsorption reacts antigen-specific. The implications of the method are discussed.
Zusammenfassung
Nach dem Prinzip der Immunadhärenz-Reaktion wurde eine Methode ausgearbeitet, die einen Nachweis von virusspezifischen oder zellspezifischen Antigenen auf oder in den Membranen von lebenden Zellen in vitro gestattet: Immunadhärenz-Hämadsorption.
Im Verlauf der Virussynthese lassen sich etwa 4 Std p.i. erstmals virusspezifische Antigene in oder auf der Zellmembran von Kaninchennieren-Zellen nachweisen. Die Applikation von Actidion 2 Std p.i. verhindert diese IAH. Die 4–7 Std p.i. ablaufende Riesenzellbildung beeinflußt die Intensität der Immunadhärenz-Hämadsorption zum Nachweis von zellspezifischem Forssman-Antigen von Meerschweinchennieren-Zellen nicht. Die Immunadhärenz-Hämadsorption ist antigen-spezifisch.
