Summary
PCA-reactions are diminished by extracts of peritoneal cells from BCG-sensitized guinea pigs when injected intracutaneously as a mixture with the antibody. In contrary the PCA-reactions are enhanced by cell extracts from non sensitized animals. In this local application form the extracts of neutrophilic and mononuclear exudates are likewise effective as those of lymphocytic ones.
Intravenous administration of extracts from neutrophils and mononuclears at approximately equal parts enhances the PCA-reaction. The maximum of this enhancement was observed 2 hours after the injection of extracts from non-sensitized animals and only 24 hours after the injection of the material from BCG-sensitized guinea pigs. Extracts prepared from lymphocytic exudates obtained 72 hours after intraperitoneal injection of paraffin oil diminished the PCA-reaction up to 5 hours after administration. Afterwards there is a delayed enhancement of the PCA-reaction with a maximum at 24 hours after the injection of extracts. Cell extracts of BCG-sensitized and non-sensitized animals are comparable in this respect.
The possible role of the lymph node permeability factor (LNPF) and that of the agglutination inhibitor demonstrable in these cell extracts are discussed.
Zusammenfassung
Extrakte aus Peritonealzellen BCG-sensibilisierter Meerschweinchen dämpfen nach i.c. Injektion im Gemisch mit einem Anti BSA-Serum die PCA-Reaktion. Zellextrakte von nicht sensibilisierten Tieren verstärken die Reaktion. Diese lokalen Effekte sind Ähnlich mit Extrakten aus Zellen, die vornehmlich aus neutrophilen Leukocyten oder aus mononucleären Elementen bestanden. Die i.v.-Verabreichung von Extrakten aus neutrophilen Leukocyten und mononucleären Zellen zu etwa gleichen Teilen führt im Falle von Zellextrakten nicht sensibilisierter Tiere zu einer Verstärkung der PCA-Reaktion mit einem Maximum 2 Std nach der Injektion. Bei Verwendung von Zellextrakten BCG-sensibilisierter Tiere tritt die Verstärkung verzögert mit einem Maximum 24 Std nach der Injektion auf. Bei Verwendung von Extrakten aus Peritonealzellen, die 72 Std nach i.p. Paraffin-Injektion gewonnen wurden und vornehmlich aus Lymphocyten bestanden, ist bis zu 5 Std nach der Injektion eine Dämpfung der PCA-Reaktion zu beobachten. Eine Verstärkung tritt wieder erst nach 24 Std ein. Die Zellextrakte von BCG-sensibilisierten Tieren sind in dieser Hinsicht vergleichbar.
Die mögliche Bedeutung des in den Extrakten vorhandenen Lymphknoten-Permeabilitäts-Faktors (LNPF) und des Agglutinations-Inhibitors (AI) wird diskutiert.
