Zusammenfassung
Die Bildung eines spezifischen Antikörper-analogen Produktes (AAP) wurde im zellfreien und cellulären System untersucht. Im zellfreien System wurden drei Antigene gleichzeitig zur Stimulation der AAP-Synthese verwendet. Dabei wurde keine Konkurrenz der Antigene beobachtet. Aus dieser Beobachtung wurde der Schluß gezogen, daß für jedes spezifische AAP ein besonderes Struktur-Gen vorhanden sein muß. Diese Ansicht wurde durch zusätzliche Experimente gestützt, in denen die Synthese von RNS während der AAP-Synthese gemessen wurde.
Es konnte gezeigt werden, daß in einem cellulären System zwei aufeinanderfolgende Stimulationen mit verschiedenen Antigenen gesetzt werden können; beide Stimulationen führen zur Bildung von spezifischen AAP. Die Zellkultur ist aber nicht mehr in der Lage, auf den zweiten Antigen-Stimulus zu reagieren, wenn die Zellen nach dem ersten Antigen-Stimulus mit einem Überschuß an Antigen in Gegenwart von Komplement behandelt werden. Auf Grund dieses Experimentes wird geschlossen, daß eine kompetente Zelle im Hinblick auf ihre Fähigkeit AAP zu bilden für mehr als eine Antigenspezifität zuständig ist.
Summary
The formation of a specific Antibody-analogue Product (AAP) was studied in a cell free system and also in a cellular system. Stimulation of a cell free system can be performed by combining three distinct antigens. Since no antigenic competition was observed, the conclusion is made, that each specifity requires the informative action of a special structure gen. This conclusion is supported by additional experiments in which the RNS formation rate was studied during the synthesis of the AAP. It is demonstrated that in a cellular system two consecutive stimulations can be applied with different antigens. The responsivene s of a cell to the second antigenic stimulus is abolished if complement and an excess of antigen is applied to the cells after the first antigenic stimulus. At the basis of this experiment the conclusion is made, that the responsiveness of the competent cell is multiple with respect to the formation of AAP.
