Zusammenfassung
Ein zellfreies in vitro-System wurde von Peritonealexsudatzellen hergestellt. Dieses System ist in der Lage, ein Antikörper-analoges Produkt (AAP) nach der Stimulation mit Phagenreceptorpartikeln von E. coli und Serratia marcescens zu synthetisieren. Das AAP kann nicht von demjenigen unterschieden werden, welches in einem cellulären in vitro-System gebildet wird. Die Aktivität des zellfreien synthetisierten AAP konnte durch seinen spezifisch blockierenden Effekt auf die phageninaktivierende Potenz homologer Receptor-Partikel nachgewiesen werden. Für die Bildung von AAP ist sowohl die Ribosomenfraktion, als auch der sogenannte 100000×g-Überstand notwendig. Die AAP-Synthese kann durch Behandlung des Systems mit DNase oder RNase verhindert werden. Die Gegenwart von Actinomycin und Puromycin hemmen ebenfalls die Bildung von AAP.14C-markierte Aminosäuren werden in das AAP-Molekül eingebaut.
Summary
A cell free in-vitro system, made from peritoneal exudate cells of mice, is able to synthesize an antibody-analogue product (AAP) after stimulation with phage receptor particles of E. coli and Serratia marcescens. The AAP cannot be distinguished from that synthesized in a cellular in-vitro-system. The activity of the AAP can be demonstrated by its specific blocking effect upon the phage inactivating potency of the homologous receptor particles. The ribosomal fraction and the 100.000 g supernatant are both essential for the formation of AAP. The AAP synthesis is abolished by a previous treatment of the system with DNase or RNase, and also by the presence of Actinomycin and Puromycin. The AAP synthesis cannot be prevented by adding Mitomycin to the system.14C-labelled amino acids are incorporated into the AAP molecule.
