Zusammenfassung
Peritoneal-Exsudatzellen von Mäusen wurden in vitro mit Phagen-Receptor-Partikeln gramnegativer Stäbchen stimuliert. Diese Stimulation brachte die Bildung eines Faktors in Gang, der im Extrakt der Zellen durch seinen blockierenden Effekt auf die phageninaktivierende Potenz der zur Stimulation verwendeten Receptor-Partikel nachgewiesen werden konnte. Das blockierende Prinzip erschien in den Zellen schon 10 min nach dem antigenen Reiz. Es konnte nie in der überstehenden Gewebeflüssigkeit nachgewiesen werden. Die blockierende Aktivität wanderte mit den schnellen 19 S-Globulinen. Der blockierende Effekt war spezifisch für einen bestimmten Receptortyp. Die Receptor-Partikel gingen eine feste Bindung mit dem homologen blockierenden Faktor ein. Der resultierende inaktive Komplex bindet Komplement. Der Komplex konnte wieder getrennt werden; nach der Trennung wurde voll aktives Receptor-Material und ebenso aktiv blockierendes Prinzip gefunden. Da der blockierende Faktor in vieler Hinsicht gleiche Eigenschaften wie authentische Antikörper hat, wurde er vorläufig als Antikörper-analoges Produkt (AAP) bezeichnet. Es wird angenommen, daß der stimulierende Effekt der Receptor-Partikel auf deren antigener Eigenschaft beruht.
Summary
Peritoneal exudate cells of mice can be stimulated in vitro by phage receptor particles obtained from gram negative rods. The stimulation initiates the production of a factor which can be detected in cellular extracts by its blocking effect upon the phage inactivating capacity of the receptor used for stimulation. The blocking principle appears in the cells about 10 min after the application of the receptor stimulus; it cannot be detected in the supernatant of the stimulated cell culture. The blocking activity is associated with the fast moving 19 S globulins. The blocking effect is specific for a given type of receptor. The interaction between receptor and its homologous blocking factor leads to the physical union of both reaction partners. The resulting inactive complex fixes complement; the complex is dissociable; its splitting results into full recovery of the receptor material and of the blocking principle, both being obtained in their biologically active form. Since the blocking factor exhibits in many instances the same properties as antibodies obtained by immunizing rabbits with receptor material it is provisionally designated as “antibody-analogue product” (AAP); the stimulating effect of the receptor material is considered to be bound to its antigenic properties.
