Summary
The first 10 millimetres of root tips from pea variety ‘Maiperle’ were investigated both from seedlings of different age and from excised roots cultivatedin vitro. The topic was to comprehend the difference in development between isolated roots in synthetic nutrient and normal seedling roorts. Fresh weight, cell numbers and protein content of root segments each one millimetre in length were analysed. During the first four days of culture in vitro all characteristics mentioned show distinctive changes in the individual root segments. This enables to conclude that after isolation of roots till the third day of artificial culture the elongation zone is put onward more and more toward the meristem, i. e. till this time the elongation growth of root cell is accelerated in comparison to meristem activity. From the third to the fourth day of culture retrograde changes set in. Thereby between meristematic growth and both elongation and differentiation of root cells a new equilibrium will be reached corresponding now with the conditions of culture in vitro.
