Abstract
In the cortex and medulla of rat kidney O2 pressure was polarographically measured in-vivo at normal oxygen concentration (FI O a=0.21) and in hyperoxia (FI O 2=1.0) by means of gold microelectrodes (tip diameter: 3–8 µm).
During the measurements the electrodes were gradually moved from the cortex to the apices of the renal pyramids. The PO 2 distribution in the cortex and medulla were recorded, PO 2 histograms and mean tissue PO 2 determined. In control experiments, renal blood flow, the blood gases and renal O2 supply were measured.
Evaluation of the measured PO 2 profiles in normal oxygenation and hyperoxia shows 2 zones of different O2 supply. The zone of high PO 2 values lies in the cortex and outer medulla with a mean O2 pressure of 38 mm Hg (normal) and 77 mm Hg (hyperoxia). The zone of low PO 2 values is restricted to the inner medulla and papilla with a mean PO 2 of 9 mm Hg (normal) and 18 mm Hg (hyperoxia) respectively. In the region between outer and inner medulla we found a typical tissue PO 2 decrease about 30 mm Hg. This phenomenon must be attributed to an O2 diffusion shunt in the vasa recta.
Zusammenfassung
Unter standardisierten Bedingungen werdenin vivo die O2-Drucke unter Normoxie (FI O 2=0,21) und Hyperoxie (FI O 2=1,0) in Rinde und Mark der Rattenniere polarographisch mit Goldmikroelektroden (Spitzendurchmesser 3–8 µm) gemessen. Bei kontinuierlichem Vorschub der Elektrode wird diepo 2-Verteilung in Rinde und Mark ermittelt,po 2-Histogramme aufgestellt und die mittleren Gewebs-po 2-Werte errechnet. In Kontrolluntersuchungen erfolgte die Messung der Durchblutung, der Blutgasdaten und der O2-Aufnahme der Niere bei Luft- und O2-Atmung.
Das Ergebnis derpo 2-Messungen im Nierengewebe unter Normoxie- und Hyperoxiebedingungen zeigt, daß die Niere in eine Zone hoher und eine Zone niedriger O2-Drucke gegliedert ist. Im Bereich der Cortex und äußeren Medulla beträgt der mittlere Gewebs-po 2 unter Normoxie 38 mm Hg und unter Hyperoxie 77 mm Hg; in der inneren Zone und in der Papille liegt der mittlerepo 2 unter Normoxie bei 9 mm Hg und unter Hyperoxie bei 18 mm Hg. Der typische Gewebs-po 2-Abfall um ca. 30 mm Hg am Übergang der äußeren in die innere Zone der Medulla wird durch einen O2-Diffusionsshunt in der vasa recta bedingt.
