Zusammenfassung
Von steril angezogenen, 3 Tage alten Erbsenkeimlingen wurden Wurzelkulturen in einer modifizierten Nährlösung nach Bonner und Addicott ohne und mit Zusatz von 2 · 10−8 M/l Dichlorphenoxyessigsäure (2,4-D) angelegt. Das Wachstum von Keimlingswurzeln, normalen und 2,4-D-Kulturwurzeln wurde quantitativ analysiert. Die von Brown und Wightman (1952) gefundene Abhängigkeit des Wachstums von der Ausgangslänge der isolierten Wurzeln gilt auch noch für über 10 mm lange Wurzelspitzen. Durch 2,4-D werden Erbsenwurzeln in Kultur viel stärker beeinflußt als Maiswurzeln (Kandler 1950); in Lösungen mit 1 · 10−7 M/l 2,4-D wachsen Erbsenwurzeln nicht mehr.
Die ersten fünf, jeweils 2 mm langen Segmente der drei Wurzeltypen wurden durch Zellenzahl, Frischgewicht, Trockengewicht, Wassergehalt und Gehalt an ‘Protein’-Stickstoff (PN) charakterisiert. Bei Kultur in vitro wird die Entwicklung der Wurzeln in bestimmter Weise verändert, vor allem werden die Differenzierungsvorgänge beschleunigt; in besonderem Maße ist dieses in 2,4-D-haltiger Nährlösung der Fall. Dabei ist der Unterschied zwischen Keimlingswurzeln und Kulturwurzeln größer als zwischen den beiden Arten von Kulturwurzeln (ohne und mit 2,4-D).
Nach Übertragung der in 2 mm lange Segmente zerteilten Wurzeln in saccharosehaltige Lösung nimmt die Trockenmasse gewöhnlich zu, das Spitzensegment mit dem Wurzelmeristem ausgenommen, das an Trockenmasse verliert. In anorganischer und in citrathaltiger Lösung tritt stets ein Gewichtsverlust ein.
Der Sauerstoff-Verbrauch der verschiedenen Wurzeltypen ergibt je nach der Bezugsgröße ein unterschiedliches Bild. Im allgemeinen folgt der in 8 Stunden gemessene O2-Verbrauch, bezogen auf gleiche Stückzahl, den Veränderungen des Trockengewichtes in dem zur Atmungsmessung benutzten Medium, z. B. steigt die O2-Aufnahme in Sacharose-Lösung im allgemeinen an, doch kommen Ausnahmen von dieser Regel vor. Bezogen auf gleiche Zellenzahl, nimmt der O2-Verbrauch in den meisten Fällen vom ersten Segment an laufend zu, bezogen auf PN, ergibt sich ein Maximum in einem bestimmten Segment, das bei den drei Wurzeltypen durch das erste Auftreten von Xylemelementen gekennzeichnet ist. Bei Kulturwurzeln ist es das zweite (2–4 mm), bei Keimlingswurzeln das vierte (6–8 mm) Segment.
Summary
Excised roots of 3 days old pea seedlings were cultivated in sterile nutrient solutions (Bonner and Addicott) with and without addition of 2 · 10−8 M/l dichlorophenoxyacetic acid (2,4-D). Quantitative analysis of growth of seedling roots and roots from normal and 2,4-D solutions revealed that relations between elongation and initial length of isolated roots, as found by Brown and Wightman (1952), are valid even for root tips longer than 10 mm. Cultivated pea roots being much more influenced by 2,4-D than maize roots (Kandler 1950) do not exhibit any growth in solutions with 1 · 10−7 M/l 2,4-D.
The first five segments of 2 mm length from the above named three root types were defined by number of cells, fresh-weight, dry-weight, water content, and content of ‘protein’ nitrogen (PN). Cultivation in vitro changes the development of roots accelerating first of all the processes of differentiation, in particular in solutions with 2,4-D. The difference between seedling roots and cultivated roots is greater than that between both types of cultivated roots (with and without 2,4-D).
Dry matter of roots dissected in 2 mm long segments ordinarily increases after translocation into sucrose solutions, except the tip segments with root meristem which looses dry-weight. Loss of dry-weight always happens in inorganic and citrate solutions.
Oxygen is consumed in different manner by the different root types, results depending from relation system. O2-uptake measured over 8 hours, if related to equal number of segments, in general follows the changes of dry-weight in the medium used for respiration measurement; e.g. in sucrose solutions O2-uptake generally increases exceptions, however, may occur. If related to equal number of cells, O2-consumption in most cases increases continuously from the first segment on; if related to PN, a maximum occurs in a certain segment being defined by the first appearance of xylem elements, i. e. the second segment in cultivated roots (2–4 mm), the fourth one (6–8 mm) in seedling roots.

Семена гороха проращивались в стерильных условиях. От проростков в трехдневном возрасте закладывались культуры корешков в модифицированном растворе Боннера и Эддикота без и с прибавлением дихлорфенокси-ук сусной кислоты (2,4-Д) в концентрации 2 · 10−8 М/л. Рост корешков количественно анализировался в обоих вариантах опыта. Зависимость роста от исходной длины корешков (Brown and Wightman 1952) действительна еще и при длине верхушек в 10мм. Корешки гороха угнетаются в культуре 2,4-Д значительно сильнее, чем корешки кукурузы (Kandler 1950); при концентрации 1 · 10−7 М/л они уже не растут вовсе.
Первые 5 сегментов (каждый длиной в 2мм) трех типов корешков характеризуются числом клеток, сырым и сухим весом, содержанием воды и «протеинного» азота. В культуре in vitro развитие корешков изменяется в определенном направлении; главным образом ускоряются процессы дифференциации; это особенно заметно в питательном растворе, содержащем 2,4-Д. Разница между контролем и культурами оказалась при этом больше, чем между обоими вариантами культур.
При переносе сегментов корешков (2мм длины) в раствор сахарозы обыкновенно наблюдается прирост сухого вещества; это правило не действительно для верхушечного сегмента, сухое вещество которого уменьшается. В неорганических растворах и в растворах, содержащих цитраты всегда наблюдается потеря в весе.
Потребление кислорода различными типами корешков дает различные значения в зависимости от способа учета. В общем потребление O2, измеряемое в течение 8 часов и приведенное к равному числу сегментов, следует изменениям сухого веса в среде, которая употреблялась для измерения дыхания; так напр. потребление O2 в растворе сахарозы в общем повышается, но встречаются и исключения из этого правила. Потребление O2, приведенное к равному числу клеток, в большинстве случаев, увеличивается начиная с шервого сегмента. Если делать учет по «протеинноме» азоту, то максимум потребления обнаруживается в определенном сегменте, который у всех трех типов корешков характеризуется первым появлением элементов ксилемы. У корешков в культуре таковым является второй сегмент (2–4мм), у контроля — четвертый (6–8мм).
